Objective: Study on the correlation between anti-anxiety effects andchanges of pharmaceutical chemical composition in cerebrospinal fluid ofSuanzaoren Decoction in Medical Treasures of the Golden Chamber, providesthe basis for the research on material basis of anti-anxiety effect, mechanismand the research and development of new drug for anti-anxiety effect. Basedon the above results of research, we carried on the preliminary explorationof thought and the method about researching on "Chinese medicine compound CSFdrug chemical".Methods:①Detecting volatile ingredients of Suanzaoren Decoction and itssingle herbs by gas chromatography-mass spectrometry (GC-MS), comparingsimilarities and differences of chemical composition from the whole party andits single drug;②Testing the differences among medicine chemicalcomposition in fingerprint of chemical composition of Suanzaoren Decoctionand its single herbs, medicated serum of Suanzaoren Decoction and medicatedcerebrospinal fluid of Suanzaoren Decoction, comparing the similarities byhigh performance liquid chromatography (HPLC), and observe the difference of chemical composition of medicated serum medicated and the medicatedcerebrospinal fluid in different dosage of Suanzaoren Decoction;③Preliminaryly identifying part of chemical composition of the medicated serumand cerebrospinal medicated fluid of Suanzaoren Decoction by high performanceliquid chromatography-mass spectrometry (HPLC-MS);④Enzyme-linkedimmunoassay is used to observe the effect of different doses of SuanzaorenDecoction on mRNA expression of GABA alpha1receptor in cerebral cortex ofthe rabbit and content of noradrenergic(NE) in cerebral hippocampus of rabbit.Results:①In all volatile components of Suanzaoren Decoction,20compounds are found, those do not belong to any one of single herbs, of whichcontent of1-phenyl-1-pentanone is the highest,(S)–1–methyl–4–5–methyl-4–methyl-1-hexene base)-cyclohexene take second place;②In fingerprint of medicated serum of Suanzaoren Decoction, there are twoserum natural ingredients, four drug-induced constituents, one metabolite;In fingerprint of medicated CSF of Suanzaoren Decoction, there are two serumnatural ingredients, four drug-induced constituents, one metabolite; and withthe increase of dosage, content of parts chemical component continues toincrease, parts appear at large doses, parts continues to decline, Some firstincreased then decreased, content reached the highest at high doses;③Preliminaryly identifying by HPLC-MS, six drug-induced constituents infingerprint of medicated serum of Suanzaoren Decoction contains Spinosin,ononin, Timosaponin EI, Jujuboside BI, four drug-induced constituents infingerprint of cerebrospinal fluid of Suanzaoren Decoction contains Spinosin,ononin, Timosaponin BII;④With the increasing of the dosage, mRNA expressionof GABA alpha1receptor in cerebral cortex of the rabbit increased and contentof NE in cerebral hippocampus of rabbit is in the fall.Conclusion:①20compounds are found in all volatile components,whichdo not belong to any one single herbs of the decoction, speculates they areproducts after decocting of Suanzaoren Decoction;②Spinosin, Jujuboside BI, ononin, Timosaponin EI and No.5in medicated serum, the above5kinds ofcompounds are the most easy to work into the blood. While Spinosin, TimosaponinBII, ononin, that three kinds of compounds are the drug chemicals that onesthe most easily go through the blood brain barrier and get into thecerebrospinal fluid, they may play central utility on efficacy material baseof Suanzaoren Decoction. According to comparison of content of component inthe medicated serum and cerebrospinal fluid, component of Zizyphus jujubebenevolence is easier to go into the cerebrospinal fluid to exert its effect;③Spinosin, ononin, Timosaponin BII in the cerebrospinal fluid may be efficacymaterial base of Suanzaoren Decoction in centre. Provides the basis for theresearch on material basis of anti-anxiety effect, mechanism and the researchand development of new drug for anti-anxiety effect, provides new ideas andmethods for the research on central material basic to search on the restlessclass of traditional Chinese medicine compound.
