objective:By observing the Zhilong Huoxue Tongyu capsule on endothelial function and angiogenesis of myocardial infarction rats, to explore Zhilong Huoxue Tongyu capsule on myocardial infarction in the rat model of myocardial protection of the possible mechanism.Methods:By ligating the left anterior descending artery of coronary artery of myocardial infarction model of rat. Health and SD,36male rats were randomly divided into sham group, model group, Low-dose group of Zhilong Huoxue Tongluo capsule (the low dose group for short), Zhilong Huoxue Tongluo capsule middle-dose group (the middle dose group for short), Zhilong Huoxue Tongluo capsule high dose group (the high dose group for short), tongxinluo group,6rats in each group. In sham group and model group,rats saline lavage daily, the other group were given corresponding drug lavage for14days. In postoperative of7and14days,detection of serum levels of carbon monoxide synthase (eNOS), endothelin-1(ET-1),von willebrand factor (vWF), vascular endothelial growth factor (VEGF),14days after death in heart tissue of rats line HE dyeing infarction area morphology observation, immunohistochemical method can detect micro vascular edge area of infarction (MVC).Results:The7th day, compared with sham group, group of eNOS, ET-1, vWF and VEGF are higher, the difference was statistically significant (P<0.05). Compared with model group, VEGF and eNOS in each treatment group were higher (P<0.05), of which the high dose group and tongxinluo group increased significantly,have a significant difference (P<0.01); ET-1and vWF treatment group were decreased, the high dose group and tongxinluo group decreased significantly, have a significant difference (P<0.01). Compared with tongxinluo group, high dose group of each index had no significant difference (P>0.05). The14th day, compared with the sham group, other group of eNOS, ET-1, vWF and VEGF were higher, the difference was statistically significant (P<0.05), compared with model group, VEGF and eNOS in each treatment group were higher (P<0.05), of which the high dose group and tongxinluo group increased significantly,have a significant difference (P<0.01). ET-1and vWF in each treatment group were decreased, the high dose group and tongxinluo group decreased significantly,have a significant difference (P<0.01), compared with tongxinluo group, high dose group of each index had no significant difference (P>0.05). Compared with model group,infarct size (IS) of each dosage group were lower, middle dose group,high dose group and tongxinluo group had significant difference (P<0.01), high dose group and tongxinluo group has no statistical significance (P>0.05). MVC counting results:compared with sham group, the other group had significant changes (P<0.05), compared with model group, the MVC treatment group were higher (P<0.05), high dose group and tongxinluo group increased obviously, there are significant difference statistically significant (P<0.01), compared with tongxinluo group, high dose group of MVC there was no statistically significant difference(P>0.05). Conclusion:1. Zhilong HuoxueTongyu capsule have protective effect to myocardial cells on myocardial infarction rats and reduce infarction size.2. The mechanism that is Zhilong HuoxueTongyu capsule on myocardial infarction of the rat myocardial cells have a protective effect, may be activate eNOS、VEGF, inhibition of the expression of ET-1and vWF, thus protect vascular endothelial cells, promote new blood vessels form edge area of infarction.2.The capsule can promote new vessels comformation of myocardial infarction of edge area in the rat infarction, its mechanism may be increase VEGF expression,3.The capsule has a certain concentration-response relationship on protective effect in protecting myocardial infarction of the rat endothelial cells, and comformation new blood vessels.
