Objective: This study was designed to detect the different expressionand correlation of nuclear factor-kappa B(NF-κB) and intercellular adhesionmolecule-1(ICAM-1) in the epithelium cells of different groups human anteriorlens capsule and the different expression and correlation of NF-κB P65mRNAand ICAM-1mRNA in the different groups of human lens epithelial cells, toexplore the role of NF-κB and ICAM-1and relationship btween them on theoccurrence and development of the diabetic cataract.Methods:5normal lens anterior capsules (normal control group)and30anterior capsules with age-related cataract (diabetes mellitus group) and30anterior capsules with diabetic cataract (age-related group) were collectedrandomly. The expression of NF-κB P65and ICAM-1protein were detected byimmunohistochemistry. Human lens epithelial cells were respectively culturedwith low glucose (5mM glucose), high glucose (25mM glucose) and manniol(20mM mannitol). The expression and correlation of NF-κB P65mRNA andICAM-1mRNA were detected by reverse transcription-polymerase chainreaction (RT-PCR). Image the Pro6.0professional image analysis system wasused for measuring the value of the mean optical density (MOD) in positivecells and Quantity one software was used for analyzing the gray value of PCRproducts. The statistical methods were carried out by the SPSS17.0softwareand the significant level was defined as P<0.05. Results:1. The expression value of NF-κB P65and ICAM-1in the epitheliumof human anterior lens capsule of diabetic cataract were increased obviously,compared with the age-related cataract group (P<0.01, P<0.01respectively).2. In the age-related cataract and diabetic cataract, there was a statisticallysignificant correlation between NF-κB P65and ICAM-1, with respectivelycorrelation coefficients of r=0.408, P<0.05; r=0.766, P<0.01.3. The expression value of NF-κB P65mRNA was increased obviously,compared with the normal group and mannitol group(P<0.01, P<0.01respectively), there was no statistically significant difference between thenormal group and mannitol group(P>0.05);the expression value of ICAM-1mRNA was increased obviously, compared with the normal group and mannitolgroup(P<0.01, P<0.01respectively), there was no statistically significantdifference between the normal group and mannitol group(P>0.05).4. There was a statistically significant correlation between NF-κB P65andICAM-1mRNA in the high-glucose group(r=0.892,P<0.05),but in the normalgroup and mannitol group,there were no statistically significant correlation,with respectively correlation coefficients of r=0.391, P>0.05; r=0.633, P>0.05.Conclusion:1. Compared with the age-related cataract and the control group, there was ahigher expression of NF-κBP65and ICAM-1and a statistically significantcorrelation between them in diabetic cataract. 2. High glucose can promote NF-κB P65mRNA and ICAM-1mRNAtranscription, and the effect is not due to the osmotic pressure caused by highglucose; there was a statistically significant correlation between NF-κB P65mRNA and ICAM-1mRNA in the high glucose condition.3. NF-κB P65may affect the occurrence and development of diabetic cataractby regulating ICAM-1.
