ObjectiveTo investigate the association of angiotensin Ⅱ type Ⅰreceptor (AT1R)gene polymorphism with AT1R expression levels and brain edema inhypertensive intracerebral hemorrhage.Methods45patients of hypertensive intracerebral hemorrhage undergoingoperations were collected from December,2011to August,2012in theNeurosurgery of Yongchuan Hospital, Chongqing Medical University:1.The AT1R A1166C polymorphism was assayed by restrictionfragment length polymorphism (RFLP) analysis;2.Brain tissue was taken from lesions in operation, then AT1R mRNAconcentration was determined by RT-PCR and AT1R protein level wasdetermined by Western blot; 3.Cerebral edema was evaluated by a non-invasive brain edemamonitor at24h,48h and72h after operations.Results1.There were36cases of AA genotype,9cases of AC genotype, andthe frequency of A and C alleles was90%and10%respectively;2.The concentration of AC genotype AT1RmRNA were slightly higherthan the AA genotype(P=0.52), but the difference was not statisticallysignificant;3. The concentration of AC genotypes AT1R protein in brain tissuewas significantly higher than the AA genotype (P=0.011);4. No significant differences were found in the cerebral edema factorsbetween the two genotypes within24hours (P=0.226), but significantdifferences were found in48hours (P=0.034) and72hours (P=0.005).Cerebral edema factor in the AC genotype is higher than the AA genotype.Conclusion:1.There was no significant correlation between the AT1R genepolymorphism and AT1RmRNA concentration in the brain tissue;2.There were significant correlations between AT1R genepolymorphism and AT1R protein concentration in the brain tissue, AT1Rprotein concentration of the C allele carriers was higher;3.There were significant correlations between AT1R genepolymorphism and the extent of cerebral edema in hypertensive cerebral hemorrhage, the more frequent occurrence of cerebral edema were foundin the C allele carriers.
