Objective(1)Establish rabbit skeletal muscle contusion injury model.(2) HEstaining and Ultrasound imaging were employed to inspect pathologicalchanges and effect of massage on blood flow and vascular repair in theprocess of skeletal muscle recovery.(3) To investigate the effect of massageon growth factor bFGF and IGF-Ⅰin the process of Skeletal musclerecovery.(4) Based on the skeletal muscle recovery process of TGF-β1mRNA and COL-Ⅰ mRNA relative expression, to investigate the effects ofmassage on the formation mechanism of inhibiting scar tissue of skeletalmuscle.MethodsThirty-two New Zealand white rabbits, weighing (2.0±0.5) kg, Afteradaptive feeding for7d were randomly divided into4groups: normalcontrol (A)(n=4); the control group before the massage (B)(n=4);self-repairing group (C)(n=12,further divided into7d,14d and21dtime points), and massage group(D)(n=12,further divided into7d,14d and 21d time points). In group A, the rabbits were not injured as normalcontrols, in groups B、C and D, the rabbit models of quadriceps femorisinjury were prepared by self-made beater. In group C, no massage therapywas given as natural recovery controls; in group D, massage therapy wasgiven after5days since model built successful as massage group. C and Dgroup after injury in7d,14d and21d each take4different animalsquadriceps samples. HE staining were employed to inspect pathologicalchanges, Ultrasound imaging was employed to inspect the tissue necrosissituation of7d、14d groups after building the model. Immunohistochemicaldetection of the expression of bFGF protein and IGF-Ⅰprotein, The SYBRGreen real-time quantitative RT-PCR method was employed todetectIGF-ⅠRNA、 TGF-β1RNA and COL-ⅠRNA expressions.ResultsAfter massage intervention, Ultrasound of micro-bubble angiographyand HE staining showed that compare with the unmassaged groups (C),themassaged groups(D) injury parts is lighter, blood supply is richer, necroticarea is significantly decreased, and vascular restore faster. the expression ofbFGF in massage group> natural recovery group in7d groups(P﹤0.05),and in14d、21d groups natural recovery group>massage group(P﹤0.05).IGF-Ⅰ protein and mRNA expression the massage group is higher thanthe natural recovery group in About7days(P﹤0.05), and in14d、21dgroups natural recovery group>massage group(P﹤0.05). The TGF-β1 mRNA expression of the volume,the experimental group and control grouphad no significant difference in7d group (P﹥0.05), the expression ofTGF-β1mRNA were significantly less than the amount in control group ingroups14d and21d (P <0.01). The COL-ⅠmRNA expression of thevolume,the experimental group and control group had no significantdifference in7d group (P﹥0.05), the expression of COL-Ⅰ mRNAwere significantly less than the amount in control group in groups14d and21d (P <0.01).ConclusionMassage can effectively prevent muscle atrophy, degeneration andnecrosis, provide a rich blood supply for the damage regions, promote theangiogenesis, and significantly improve the histomorphology recovery. Themechanism may be through raising the IGF-I and bFGF to promote skeletalmuscle tissue repair in early injury. At the same time massage caneffectively reduce TGF-β1mRNA and COL-Ⅰ mRNA expressions,reduce scar formation in the process of dental repair in order to achievepromote the function recovery purpose.
