Some researches speculate that the increased ability for competition may largely depend on the functions of skeletal muscles. As a result, an increasing attention has been paid to the studies on skeletal muscles. It is known that long time training, ischemia, hypoxia may have biological compensatory effects on histology, blood kinetics, and substrate metabolizing that enhances oxygen transport, substrate oxidation and tissue growth. The angiogenesis of capillary is an important pattern of compensation in skeletal muscles, which would alleviate the limitation of tissue oxygen diffusion. This produces the increase of muscular oxygen consumption, which improves one's performance under hypoxic conditions. So the angiogenesis of capillary contributes one possible type of adaptation to the hypoxic condition in skeletal muscles.However, whether long time training promotes the growth of capillary remains controversy in the literature. Though some researchers support that training would produce greater capillary growth, they haven't got the some conclusion. There lacks conclusive evidence that the training increase mammal skeletal muscular capillary.so, we must search the mechanism,which lead to capillary growth.many factors have been found to be able to influence the formation of new capillaries, among which VEGF seems to be the most important mediator of trainig-induce capillary growth. Once,VEGF binding specific receptor FIK-1,which will have de stronger function on capillary growth.bFGF can be synthesized in endothelial cell, smooth muscle cell, adventitial fibroblasts. bFGF can improve capillary density and up-regulation effects of VEGF Gene Expression. The effects of different exercise on VEGF,FIK-1,bFGF mRNA expression will be different? We must have a depth research.therefore, we observed VEGF, FLK-1, bFGF mRNA expression and capillary density changes in the skeletal muscle of 10 months old C57 male mice by three different kinds of movements, in order to examine the effects of exercise on skeletal muscle protein synthesis. we purpose si to find different effects on angiogenesis and aerobic capacity by different movements, provides the instruction for sports training, physical exercise and Physical education.Experimental Methods:This study selected 32 10-month-old C57 male mice were studied and randomly divided into a quiet control group (C), endurance exercise group (E), ladder exercise group (R), mixed-exercise group (E+R), there are 8 of each. Of the selected objects were used as the running platform and ladder weight-bearing endurance training and resistance training movement patterns.28 weeks off 48 hours after the end of the exercise were sacrificed, quick to take the right posterior limb in mice gastrocnemius, weighing, numbering, placed in Eppendorf tubes in liquid nitrogen quick-freezing,-80℃refrigerator. Application of RT-PCR detection method to determineVEGF, FLK-1, bFGFmRNA expression changes, exercise, and AKP staining of frozen sections was observed related to changes in muscle capillary density.Results:After 7months training, body weight be increased in all groups of mice, control group have the greatest weight change,and the endurance exercise group have the minimum weight change. in the four groups, weight ratio of gastrocnemius/ body weight be increased. compared with the control group,VEGF,FLK-1,bFGFmRNA have nothing change in endurance exercise group, VEGF,FLK-1,bFGFmRNA significantly increase in resistance exercise group;FLK-1,bFGFmRNA significantly increase in mix-exerciseConclusion:resistance exercise will have best effect on the expressions of VEGF,FLK-1,bFGFmRNA, the resistance exercise will improve the function of endothelial cell, lead to influence the formation of new capillaries which can improve the function of muscle, it is the besy way for body building. mixed-exercise effect on the expressions of FLK-1,bFGFmRNA,it can be a better way for body building, endurance exercise can't effect on the expressions of VEGF,FLK-1,bFGFmRNA,but it can inprovr the new capillaries,we need forther research to find the machinsm.
