ALD and NAFLD existed to the risk of liver cirrhosis, liver cancer, liver failure, has become one of the world’s important public health problem in the21st century. The first part of this study were collected the huaian city for retirees of enterprises and institutions of the physical examination data, and obtained the certain people of ALD and NAFLD prevalence.The exact pathogenesis of NAFLD is unclear. miR-143is the first discovered with NAFLD related miR. Studies have shown that, miR-143involved in regulating the body’s fat metabolism. Therefor, the second party of this study discusses the regulation of miR-143in HepG2cells lipid metabolism. Section oneObjective:To investigate the prevalence and major risk factors of fatty liver disease (FLD) in Huaian city for retirees of enterprises and institutions.Methods:The physical examination data on13112retirees were obtained from Healt-h Examination Center of the82nd Hospital, from September,2011to December,2012.Results:1.5853people were detected with FLD, and the total prevalence rate was44.63%, while female prevalence was46.62%higher than that of male(41.33%) with a significant difference;2. FLD group of four types of hyperlipidemia, impaired fasting glucose, diabetes, hypertension were significantly higher than in non FLD;3. Multiple Logistic regression analysis revealed that risk factors of FLD included sex(female), alanine aminotransferase, impaired fasting glucose, hyperglycaemia, triglyceride, low density lipoprotein cholesterol, hypertension.Conclusion:Huaian city for retirees of enterprises and institutions, the prevalence of FLD is higher, and coexist with many other metabolic diseases, we must strengthen comprehensive prevention and control. Objective:Explore the regulation of miR-143on lipid metabolism in HepG2cells.Methods:In vitro cultivation of HepG2cells. The culture medium was added FFA, while miR-143transfection and control, using qRT-PCR technique to detect the situation HepG2cells transfected with miR-143’s. Oil red O staining situation lipid accumulation in HepG2cells. Using lipoprotein lipase-glycerol phosphate oxidase-peroxidase4-amino antipyrine and phenol method assay in HepG2cells on the Olympus AU640automatic biochemical analyzer TG content. Results:The tendency for0.25mmol/L FFA can result in HepG2TG significantly ac-cumulated in the cell, and the time dependence. miR-143expression can significantly inhibits lipid accumulation in HepG2high FFA condition.Conclusion:miR-143may be involved in the obese state of lipid metabolism in liver cells regulation.
