Diarrhea is a independent disease caused by different kinds of pathogen and reason, and accompanied with other disease. Recently, problems of abusing of antibiotics, environmental pollution, and food safety make diarrhea more frequent, which has become the second motality reason of children below five years old. More and more medical workers consider that the plant resources of our country can replace antibiotics andavoid side effects aroused by synthetic drugs. Fuzhuan tea is an antidiarrhea plant drink in our province, which can effectively treat many sorts of diarrhea including secretory diarrhea, permeability diarrhea, and exudative diarrhea. But, the mechanism of antidiarrhea is sill unclear. To explicit the anti secretory diarrhea mechanisms of Fuzhuan tea, the effect of Fuzhan tea on gut microbes, gut mucosa, inflammatory cytokines5-HT, and the expression of NHE on gene and protein level were explored by establish seccretory diarrhea mouse mode by Cassia angustifolia Vahl.By system research, the reasluts received as below:1. Cassia angustifolia Vahl induced mice diarrhea and reduced beneficial micro-organismsThis research showed that Cassia angustifolia Vahl not only caused diarrhea of mice, but also resulted in harmful intestinal microbial’s proliferation, beneficial microorganisms’decreasing, intestinal mucosa structure’damaging, the5-HT of secrum and intestinal tract’s increasing, and the expression level of NHE-3protein’s rise.Therefore, Diarrhea caused by Cassia angustifolia Vahl was not only the secretory diarrhea.2. The precaution and treatment effects of Fuzhuan tea on diarrhea caused by Cassia angustifolia VahlThe Fuzhuan tea could precaute diarrhea caused by Cassia angustifolia Vahl, which obviously reduced loose stool rates but not diarrhea index. Fuzhuan tea could strengthen the immunity of mouse, maybe because of it contain many active ingredients, such as polyphenols, caffeine, polysaccharide and so on.It could prevent mouse from diarrhea. 3. Fuzhuan tea regulate intestinal flora disturbance caused by Cassia angustifolia VahlIn vitro tests results showed that Fuzhuan tea has certain inhibitory effect on Bifidobacterium spp, Lactobacillus spp, Enterococcus and E. coli, but little inhibitory for Lactobacillus spp, Bifidobacterium spp. Daily tea drinking would not directly kill beneficial gut microbes. Fuzhuan tea had strong inhibitory effect on a variety of pathogenic bacteria, and weak effect on Bifidobacterium spp and Lactobacillus spp. In vivo tests showed that Fuzhuan tea promoted beneficial microorganisms especially Lactobacillus spp, and inhibited harmful microbes. The proliferation of Lactobacillus spp dose related to Fuzhuan tea’s dose. Fuzhuan’s inhibitory effect on beneficial gut microbes was worse than berberine and Cassia angustifolia Vahl, while better effect on harmful bacteria in high dose, which is accord with vitro resluts. It concluded that Fuzhuan tea could regulate intestinal microbes and intestinal flora disturbance caused by senna.4. The precaution and renovation Fuzhuan tea on intestinal mucosa’ hurtRoutine pathological section was applied to observe the effect of Fuzhuan tea on intestinal morphology. The results showed that derenching Fuzhuan tea before diarrhea could relieve membrane shedding phenomenon of ileum and colon mucosa, which’s intestinal morphological structure was clear and swelling was not obvious. And the moderate dose’effect was best.Derenching after Diarrhea could ease Hairs’fall off and Epithelial cell’s necrosis caused by Cassia angustifolia Vahl, but not more obvious than precaution group. Treating diarrheal mouse with Fuzhuan tea could renovate colon mucosa’s hurt, high (210mg/mL) and moderate (105mg/mL) dose was better.5. Fuzhuan tea lower the content5-HT of serum and intestineFuzhuan tea derenched before or after dirrhea could obviously lower the content5-HT of serum and intestine, but not related with it’s dose. Fuzhuan precaution group could obviously lower5-HT’s expression of colon. Moderate dose treatment obviously brought5-HT’S expression of ileum and colon. The target positon of precaution was colon and of treatment group was ileum and colon.6. Fuzhuan tea regulate the protein and gene expression level of sodium hydrogen conversion proteinFluorescence quantitative PCR and Immuno histochemical technique was applied to research effects of Fuzhuan tea on the protein and gene expression level of sodium hydrogen conversion protein. The resuts showed that the rise gene expression of NHE-1and NHE-3caused by Cassia angustifolia Vahl was reduced by Fuzhuan tea. For precaution group’s, the mRNA expression ascended. Precaution and treatment wih moderate, low dose separatedly could reduce NHE-3expression of ileum brush border, less than high dose.High and low dose Fuzhuan tea could reduced NHE-3expression of colon brush border.It can be concluded that the target positon of Fuzhuan tea on ileum and colon was different.
