Obesity has become one of the primary health concerns in many regions of the world due to its increasing contribution to overall disease burden. It was reported that dark tea is consumed attributed to its potential weight loss functionality. Fuzhuan brick tea is one of the major brands of dark tea. Own to fungal fermentation, Fuzhuan brick tea possess a unique flavor and quality. Based on previous research, it was studied in this paper by comparison of anti-obesity of the raw dark green tea (RDGT), fuzhuan brick tea (FBT) and jinhua fungi (JF) which come from fuzhuan brick tea. The results were showed as follows:1. The effect of Fuzhuan brick tea on differentiation of murine3T3-L1preadipocytes3T3-L1preadipocytes were cultured with water-soluble extract of RDGT, FBT and JF in different dosages. Cell growth was assayed by cell counting, while cell proliferation was analyzed by methyl thiazolyl tetrazolium colorimetric assay(MTT).The degree of cell differentiation was evaluated by oil red O staining and spectrophotography. Expression mRNA levels of the C/EBPα、PPARγ2、PTP1B and GLUT4genes were evaluated by RT-qPCR. RDGT, FBT and JF had no significant influence on the proliferation and growth of3T3-L1preadipocytes, but could inhibit3T3-L1preadipocytes differentiating into mature adipocytes in a dose-dependent way. JF which inhibited3T3-L1cells fat deposition reaches67.83%at200μg/mL. All of the water-soluble extracts were can markedly increased gene expression levels of C/EBPα、PPARγ2、GLUT4compared with the control group, and markedly reduced gene expression levels of PTP1B compared with the control group.2. The effect of Fuzhuan brick tea on differentiation of Caenorhabditis elegansC.elegans was cultured with water-soluble extracts of RDGT, FBT and JF in different dosages. The degree of lipid droplets of C. elegans was evaluated by oil red O staining and spectrophotography. The result showed that200μg/mL water-soluble extracts could inhibit fat deposition the wild-type C. elegans to44.13%for JF,23.5%for FBT separately.
