| Objective This topic by establish the myocardial ischemia reperfusion induced-arrhythmia in rats by ligating left anterior descending coronary artery to studyGegenqinlian decoction in vascular and myocardial protective effects of arrhythmia inrats and to explore its mechanism.Methods①Establish the myocardial ischemia reperfusion induced-arrhythmia in ratsby ligating left anterior descending coronary artery after gavaging Gegenqinlian7d,using Ⅱ-lead electrocardiogram(ECG) measurement for recording the style and numberof arrhythmia;Determination the content of angiotensinII(AngII) in plasma byradioimmunoassay;Measuring the expression of nuclear factor-κB (NF-κB) inmyocardial tumor necrosis factor-alpha (TNF-alpha) by Enzyme-linked immunosorbentassay(ELISA);Determination the rate of myocardial infarction of model rats;②Establish the myocardial ischemia reperfusion induced-arrhythmia in rats by ligatingleft anterior descending coronary artery after gavaging Gegenqinlian7d, monitoringⅡ-lead electrocardiogram(ECG) and recording the arrhythmic continued time;Measuring the whole blood viscosity(WBV), plasma viscosity(PV) by YDA-Ⅳ BloodViscometer; Determination the content of SOD by754UV-Vis Spectrophotometer;Measuring MDA in serum by radioimmunoassay and the heart weight index;observingthe pathological changes in myocardial tissue of model rats.Results①Gegenqinlian decoction can inhibit the number of arrhythmia in model rats(P<0.05); Improve the inflammatory factors of the myocardial ischemia reperfusioninduced-arrhythmia in rats by ligating left anterior descending coronary artery: reducethe content of AngII in plasma, NF-κB in myocardial and TNF-alpha in serum(P<0.01);And reduce the rate of myocardial infarction(P<0.01);②After establishing themyocardial ischemia reperfusion induced-arrhythmia in rats by ligating left anterior descending coronary artery, the drug group could postpone and shorten the time ofarrhythmia(P<0.01) and significantly reduce blood viscosity(high,medium,low)andplasma viscosity(P<0.01); reduce the content of MDA and increased SOD activity tomaintain a balance between them(P<0.01);Compared with model group,high group andmiddle group reduce the heart weight index to inhibit cardiac hypertrophy andventricular remodeling(P<0.01,P<0.05),and to improve the pathological changes inmyocardial tissue of model rats.Conclusion①Monitoring heart fount that Gegenqinlian decoction can inhibit thenumber of arrhythmia in model rats; regulation the expression of AngII, NF-κB andTNF-alpha; reduce the rate of infarction;②Gegenqinlian could delay and shorten thetime of arrhythmias; improve hemorrheology and regulate the balance between SODand MDA,and to protect the myocardial tissue. |
