Gegenqinlian decoction is a classic traditional Chinese medicine which composed of P. Thomsonii Benth, Scutellaria Baicalensis Georgi, Coptis chinensis Franch, Glycyrrhizae, Zingiber officinale Rose. It is widely used as a treatment for anti-inflammatory and anti-virus, diarrhea, diabetes and other diseases. However, its effective components and efficacy of medicinal ways remains not yet fully clear. In this study, Gegenqinlian Decoction as the research object, by establishing cell model to explore the cell viability of aggregates which was centrifuged at 400 g and 15000 g centrifugal force and the supernatant at 400 g. The aim is to study the mechanism and pathways of Gegenqinlian decoction.1. The drug was carried through the monolayers transmembrane (AP side → BL side). The baicalin was detected by using high efficiency liquid chromatograph (HPLC) from AP and BL side. The results show that Gegenqinlian decoction is moderate absorption drug and the absorption rate between 20% and 70%. The samples of 400 g sediment,15000 g sediment and 400 g supernatant from Gegenqinlian decoction are moderate absorption of the drug, which aggregates is the best material to be absorbed.2. On the L-02 cell activity was found that Gegenqinlian decoction within a certain range of concentrations, cell viability will decrease with the decreased drug concentration and there is a dose-dependent manner. Gegenqinlian decoction is nontoxic to liver. The identification of aggregates and the supernatant from Gegenqinlian decoction activity is that aggregates is not only nontoxic to liver cells and also will promote the growth trend exhibited with protecting liver function, especially the effect of 400 g sediment.Gegenqinlian decoction can enhance the activity NR8383 cells by stimulating the cell growth. And the 15000 g sediment on cell activity is the highest of 205%.3. Detecting binding of insulin secretion, capacity generation of MDA and SOD, Gegenqinlian decoction can increase antioxidant activity of INS-1 cells which was damaged by STZ playing a protective role, while Gegenqinlian decoction also protect INS-1 cells release function. Comparing each samples of Gegenqinlian decoction precipitate aggregates showed a significant protective effect and 400 g precipitate sediment better than superior supernatant, GIS highest values of 2.74 in the sample set (control group GIS is 3.23) and β-islet cells for functional recovery has promoted. By dynamic light scattering method, the Gegenqinlian decoction 400 g,15000g sediment average particle size was detected. The results showed that 400 g and 15000 g sediment with an uneven size distribution at a range of 1484-4145nm and 295-1990 nm in diameter respectively.In summary, the aggregate of Gegenqinlian can promote absorption through the intestinal mucosa, with a pro-proliferative activity of macrophages and improve islet β cell antioxidant capacity of STZ-induced oxidative damage. However the physiological and pharmacological effects in vivo need further study.
