ObjectiveThis study is on the basis of breeding with high fat and sugar, then ratswere injected with STZ by enterocoelia to establish type Ⅱdiabetic model.Westudy themechanisms of DCM and investigate on the treatments of DCM by PC.MethodsTake Streptozotocin for diabetic model by Intraperitoneal injection. Thereare50health SD rats, Take40healthy rats gives ordinary diet after a month ofintraperitoneal injection volume and do not contain STZ of citric acid buffer, theother rats give high fat and sugar diet after a month intraperitoneal injection ofsmall doses of the chain with meal30mg urine bacteria/kg, establishtypeⅡ diabetes model.After72h, text blood sugar, blood sugar was16.7mmol/L may be set for diabetes mellitus (diabetesmellitus, DM) model success.Then randomly divided into5groups: the control group, diabetes model group,Procyanidins low (50mg/kg), medium (100mg/kg) and high dose group(200mg/kg).Procyanidins low, medium and high doses groups according to50,100,200mg/kg irrigation stomach12weeks. Through the SureStep Plusglucose monitor blood glucose level determination rats; Biochemistry examinesthe serum lipid content;CM-2000-B biomedical image analysis system detectionof rat heart function; Red meat-Iraq sappanwood dyeing the myocardial tissuepathology change; Immunohistochemistry methods were used and Western Blotprotein mark method to determine the heart tissue nuclear factor-κB (NF-κB), tumor necrosis factor (TNF-α), between the cell adhesion factor-1(ICAM-1) andmononuclear chemotaxis protein-1(MCP-1) the protein expression.ResultsCompared with control, the general situation of diabetes model rats is badand appeared more drinks, polyphagia, polyuria, weight loss and reduceautonomic activity, fur lackluster and burnout.blood sugar and blood fatincreased significantly(P<0.01),heart function decline(P<0.01), light-microscopymyocardial structure is not clear, arranging disorderly, the distortion, musclecells gap increases, inflammatory cells within the visible, myocardial tissue theNF-κB, TNF-α, ICAM-1, MCP-1the protein expression increased significantly (P<0.05); compared with diabetes model group, Procyanidins each dose group ofblood sugar and TG, TC, LDL significantly (P<0.05or P<0.01), HDL increasedsignificantly (P<0.05or P<0.01), left ventricular pressure increased rate anddrop rate of the biggest (±dp/dtm a x) are significantly increased (P<0.05or P<0.01), pathological form shows myocardial structural damage are obviouslyimprove muscle tend to tidy than the diabetes model group, structure close tonormal myocardial, did not see the inflammatory cells infiltration, myocardialtissue the NF-κB, TNF-α, ICAM-1, MCP-1the protein expression significantlyreduced, including Procyanidins high dose group is the most obvious ofimproving (P<0.01).Conclusion1、Procyanidins can alleviate DM rats pathological organizational change ofinduced by STZ, reduce blood fat and blood sugar and significantly improvediabetic rats general signs state.2、Procyanidins protects myocardial tissue of diabetic rats,it may improvediabetes rat cardiac function condition, reduce rat myocardial pathologicstructure damage, block the related expression of NF-κB、TNF-α、ICAM-1、MCP-1, thus dose dependent.
