Objective: To observe the effects of heat-sensitive moxibustion on the expression and activity of IL-1??IL-2 and TNF-? in serum and the content of NF-k B?ICAM-1 in cerebral cortex in focal cerebral ischemiarepe rfusion injury in rats,and the mechanism thereof.Methods: Eighty clean healthy male Sprague-Dawley rats,weighing 200~220 g,there were randomly divided into 3 groups: the sham-operation group,the model group and the treatment group.according to the rat tail and anal temperature changes before and behind moxibustion,the treatment group rats were divided into the moxibustion group and heat-Sensitive moxibustion group.The focal cerebral ischemia-reperfusion rat model was induced by middle cerebral artery occlusion(MCAO)for 2 h followed by reperfusion.Cerebral infarct was detected by 2,3,5-triph-enyltetrazolium chloride(TTC)staining.Brain morphology was shown by the general morphological observation and HE staining.Tail-flickla tency was determined with Tail Flick Analgesia Meter.Phlebotomizing from the abdominal aorta,mensurateing the content of IL-1?? IL-2 and TNF-? of plasma by the method of radioimmunoassay.The contents of nuclear factor-?B(NF-k B)and intercellular cell adhesion molecule-1(ICAM-1)expression in rat brain were measured by immunohistochemical methods/the immunofluorescence methods with the brain tissue paraffin/frozen section.Results: 1.Model evaluation Neural function defect scale shows the contralateral limb ischemia side appear different degree of obstacles and touch hyporeflexia.TTC staining found that the brain tissue was white in the ischemic part,lacked of typical dyeing,consistently with the middle cerebral artery distribution area.Macroscopic observation found that the general form,brain tissue of color in the ischemia side middle cerebral artery blood supply zone of rats was white,swelling obvious,brain ditch becomes shallow,the remaining parts were light red.HE staining were observed under microscope:cell lineage in the infarc tion central districts was sparse,irregular,nucleus was shrunk or dissolved,fracture.,interstitial edema;swelling of the endothelial cells.2.effect evaluation heat-sensitive moxibustion and not heat-sensitive moxibustion At after and before moxibustion,the average temperature of rat tail has no obvious change at each time in the moxibustion group,the average anus temperature at each time in the moxibustion group between before moxibustion and after moxibustion almost has no obvious change;The average tail temperature after moxibustion higher than the average tail temperature before moxibustion at each time in the heat-sensitive moxibustion group,the difference was stat istically significant(P<0.05),the average anus temperature at each time in heat-sensitive moxibustion group between before moxibustion and after moxibustion almost has no obvious change.3.Pain threshold evaluation The preoperative tail-flick latency of rats in every group has no obvious difference;The postoperative tail-flick latency of rats in the sham-operation group compared with preoperative at each time has no obvious change;The postoperative tail-flick latency of rats compared with preoperative at each time in the model group ?the moxibustion group and the heat-Sensitive moxibustion group has obvious change,there was statistically significant difference(P<0.05);In the moxibus tion group and the heat-Sensitive moxibustion group,the tail-flickl atency gradually shortened at each time after moxibustion than before,there was statistically significant difference(P<0.05);But there was no obvious change in the model group;The postoperative tail-flick latency of rats in the heat-Sensitive moxibustion group at each time are shorter,compared with in the moxibustion group.4.The content of serum IL-1 beta,IL-2 and TNF alpha in after treatment of rats In the model group rats serum IL-1 beta and IL-2 level was increa sed compared with the sham-operation group;in the moxibustion group and heat-Sensitive moxibustion group rats serum IL-1 beta and IL-2 level compared with the model group rats decreased obviously,there was statistically significant difference(P<0.05);the heat-Sensitive moxibustion group rats serum IL-1 beta and IL-2 level does not change significantly compared with the moxibustion group rats.In the model group rats serum TNF alpha level does not change significantly compared with the sham-operation group;in the moxibustion group rats serum TNF alpha level decreased slightly compared with the model group rats decreased obviously;the heat-Sensitive moxibustion group rats serum TNF alpha level compared with the moxibustion grouprats decreased obviously,there was statistically significant difference(P<0.05).5.The expression of brain tissue ischemia side of NF-k B,ICAM-1 in a fter treatment of rats In the brain tissue of the sham-operation group:the expression of ICAM-1 and NF-k Bp65 are less positive cells;expression of positive cells in the model group increased significantly than the sham-opera tion group,compared with the sham-operation group has obviously different,there was statistically significant difference(P<0.05);expr ession of positive cells in the moxibustion group and heat-Sensitive moxibustion group compared with the model group decreased significantly,compared with the model group has obviously different,there was statistically significant difference(P<0.05);expression of positive cells in heat-Sensitive moxibustion group compared with the moxibus tion group decreased,,compared with the moxibustion group has obvi ously different,there was statistically significant difference(P<0.05).Conclusion: 1.After treatment can reduce the big mouse brain tissue infarction volume,improve the movement?sensory dysfunction.2.After treatment can reduce the content of serum IL-1 beta,IL-2 an d TNF alpha and the expression of ICAM-1 and NF-k B in the brain tissue of rats,prompted the inflammation factors plays an important role in rats brain tissue injury after focal cerebral ischemia reperfusion,speculated that moxibustion therapy could suppress the expression of inflammatory cytokines,and it may be one of the im portant role in brain protection mechanism.3.The
