Object:Qingyandiwan has exactly anti-bacterial activity. However, its antiviral activity hasn’t been proved. This research is to investigate antiviral activity of qingyandiwan on common respiratory trace virus including (HSV-1), Adenovirus-11(ADV11) and influenza virus FM1(Flu).Methods:There are three parts of the experiments:The1st part is to screen antiviral activity of Qingyandiwan on HSV-1and ADV11:Here MTT colorimetric assay was used to evaluate the directly inactivate、 inhibited、treated and prevented activity. The median effective concentration (EC50) and the Treatment Index (TI) were calculated.The2ed part is to screen antiviral activity of Qingyandiwan on influenza virus FM1in mice:The influenza virus infection model was established in ICR mice to observe the following indexes:①The behavioral response and death rate of the model were observed to established the protection effect to the death of Qingyandiwan on the model.②The lung index,pulmonary pathological changes and the pulmonary virus hemagglitination titer of mice were utilized for assessment of the effects of Qingyandiwan on pneumonia of the models which caused by influenza virus FM1.③The effect on immunologic function of Qingyandiwan were detected by the spleen T and B lymphocyte transformation experiments and ELISA method.The3rd part is to screen antiviral activity of Qingyandiwan on influenza virus FM1in chick-embryo:Mix the drug with the virus for1hour then inoculate it into the chick-embryo. The hemagglutination titers of the allantoic fluids were detected.Results:1. The empirical study of antiviral activity of Qingyandiwan on HSV-1and ADV11:Qingyandiwan didn’t have treated and prevented activity on HSV-1and ADV11.However, Qingyandiwan could inactivate HSV-1and ADV11directly. The anti-HSV-1IC50was201.66μg/ml and TI was14.05. The anti-ADV11IC50was523.76μg/ml and TI was4.68. Beside, Qingyandiwan could inhibit HSV-1and ADV11.The anti-HSV-1IC50was213.39μg/ml and TI was11.49. The anti-ADV11IC50was400.88μg/ml and TI was6.69.2. The empirical study of antiviral activity of Qingyandiwan on influenza virus FM1in mice:The mortality rate of the drug group is lower then it of the model group, but it hasn’t significant difference between the two (P>0.05).However, Qingyandiwan could significantly prolong the living time (P<0.01). The results of lung index, pulmonary pathological changes and the pulmonary virus hemagglitination titer proved that Qingyandiwan had anti-inflammatory effect on pneumonia. The results of spleen index and spleen T and B lymphocyte transformation experiments proved that Qingyandiwan could promote the immunological cells proliferate and strengthening the function on immune system. The IL-10in srea increased. However,The IFN-γ、TNF-a、IL-4level decreased compared to the model group.3. The experiment on chick-embryo culture showed significantly inactivation activity on the influenza virus FM1at37℃for a hour.Conclusion:1. The empirical study of antiviral activity on HSV-1and ADV11showed that Qingyandiwan didn’t have treated and prevented activity, but could inhibit and inactivate HSV-1and ADV11directly.2. The empirical study of chick-embryo culture showed significantly inactivation activity in vitro.3. Qingyandiwan has antiviral effects on influenza virus FM1and strengthening function on immune system.
