Objective: To explore the expression of tyrosylproteinsulfotransferase,(TPST) in the alveolar epithelial tissue of normal controls and lung cancer tissue of patients with lung cancer and its clinical significance.Method:We detected the expression of TPST-1and TPST-2in the alveolar epithelial tissue of normal controls (n=50) and lung cancer tissue of patients with lung cancer (n=50) by immunohistochemical techniques, and analyzed their correlations with pathological types of lung cancer, degree of differentiation, clinical stages and lymphatic metastasis, and the correlation between C-Met and TPST. SPSS18.0was used for statistic analysis, and the statistical methods include descriptive statistics analysis, chi-square test, Fisher exact test.Results:1. The expression of TPST in the lung cancer group and normal control groupTPST-1and TPST-2in lung cancer and normal lung epithelial cells showed cytoplasmic staining of yellow, brown or tan colored particles. By immunohistochemistry in the experimental group and the control group, the expression levels of TPST-1and TPST-2were detected, the expression of TPST-1and TPST-2in lung cancer tissues were lower than that of normal control group, the difference was statistically significant (P <0.01). Within the groups, the difference between the expressions level of TPST-1and TPST-2was not statistically significant (P>0.05).2. The expression of TPST in different pathological types of lung cancerIn50cases of lung cancer patients in the experimental group were pathologically typed. We found that the Expression of TPST in adenocarcinoma、squamous cell carcinoma, and small cell lung cancer was higher.But the statistical analysis results showed the differences of the expression levels of TPST among the three pathological types were not statistically significant (P>0.05).3. The expression of TPST in different degree of differentiation of lung cancerWe assessed the degree of differentiations of the lung cancer tissues of the experimental group, and it was found that the differences of the expression levels of TPST among the three degree of differentiation groups were not statistically significant (P>0.05).4. The expression of TPST in different clinical stages of lung cancerThe positive rate of TPST-1in phase I patients was77.8%, that in phase II to IV patients were81.8%,60.9%and0%respectively, so was the expression tendency of TPST-2. There was significant difference of the positive expression rate of TPST among the four clinical stages lung cancer (P<0.05).5. The expression of TPST in lung cancer patients with or without lymph node metastasisThe positive rate of TPST in lung cancer patients without lymph node metastasis in was higher than lunge cancer patients with lymph node metastasis, there was statistically significant difference between the two groups (P<0.01), the positive rate of TPST in lung cancer patients without lymph node metastasis in lung cancer was significantly higher than the expression in TPST lung carcinoma with lymph node metastasis.6. Expression of C-Met in lung cancerWe detected the expression of C-Met and TPST parallelly in lung cancer tissues of the experimental group. A negative correlation was found between them, the higher expression level of C-Met, the lower expression level of the corresponding TPST, which was statistically significant (P<0.05).Conclusion:l.The expression levels of TPST-1and TPST-2in lung cancer tissues were lower than that of normal lung tissues, there were significant differences. But no matter in normal tissues or lung cancer tissues, there was no significant difference in the expression levels of TPST-1and TPST-2. 2. There were significant correlations between the expression of TPST-1and clinical stages, with or without lymph node metastasis. And so was TPST-2.3.No correlation was found between the expression level of TPST-1and the degree of differentiation of lung cancer. And so was TPST-2.4.The were negative correlations between the expression level of C-Met and TPST-1and TPST-2in lung cancer tissues.
