ObjectiveLung cancer is one of the highest incidence and motality in all kinds of cancer in the world. Recently, the incidence of lung cancer is rising step by step. Extensive findings suggest that heterogeneous nuclear ribonucleoprotein K (hnRNP K) is a multi-function RNA/DNA-binding protein in hnRNP family. Many research has shown that there is over expression of hnRNP K in multiplicate tumour such as rectum cancer and galactophore cancer. But the role of hnRNP K in the pathogenesis and progress of lung cancer are poorly understood. This study aims to observe the expression of hnRNP K in lung cancer tissues and normal lung tissues and explore the relationship between hnRNP K expression and the pathological types, clinical stages, immerge and metastasis in lung cancer. These will provide new experimental bases to confirm the role of hnRNP K in the pathogenesis and progress of lung cancer.MethodsThe expression of hnRNP K was studied by using the SP method of immunohistochemistry in lung tissue samples of 70 cases of lung cancer,5 cases of lung chronic inflammatory disease (pulmonary tuberculosis and bronchiectasis )and 22 cases of chest trauma. The rate of positive expression was calculated in each group and all lung caner cases were follow-up. The relationships between hnRNP K expression and the pathological types, clinical stages, the size of tumor, the degree of differentiation of tumor cells, lymph node metastasis,survival time, relapse and metastasis of post operation in lung cancer was analysed by the statistical product and services solutions (SPSS) 12.0 statistical software.Survival time were calculated using the Kaplan- Meier method and compared by the log-rank test. The P value less than 0.05 was considered significant.Results1. The expression of hnRNP K in lung cancerThe hnRNP K positive exPression was observed in the cytoplasm and nucleolus of cells presenting yellow or brown grains. No staining was observed in the normal bronchial epithelial cells in control group . The hnRNP K positive rate was respectively 54.9% in 51 NSCLC, 73.7% in 19 SCLC, 53.3% in 30 adenocarcinoma and 57.1% in 21 squamous cell cancer. Statistical analysis show that there is significantly different between neoplasm and nonneoplastic tissues (P<0.05) while hnRNP K expression was no difference in pathological types(P>0.05).There is no difference between the primary tissues and metastasis tissues(P>0.05). The intensity of hnRNP K expression is the most in the group of metastatic lymph node (P<0.05).2. Correlations between hnRNP k expressions and clinicopathlogical factors hnRNP K expression and the degree of hnRNP K expression in lung cancer was significantly associated with the size of tumor(P<0.000) and clinical stage (P<0.05) while hnRNP K expression was negatively associated with the degree of differentiation of tumor cells and lymph node metastasis.3. Correlations between hnRNP K expression and survival time,relapse, metastasis of post operation.12 of 70 cases with lung cancer lost follow. The post-operation survival time was 1-33 months at the end of this study. The mean post-operation survival period was (23.4884±1.501) months. The results show that hnRNP K expression significantly related with post-operation metastasis (P<0.05), but there is not significantly related with the intensity of hnRNP K expression (P>0.05). Statistical analysis don't show that hnRNP K exprssion relate with relapse and survival time of post operation (P>0.05).Conclusion1. hnRNP K is all over expressed in lung cancer tissues. The positive rate of hnRNP K exprssion was significantly higher than nonneoplastic tissues.2. The intensity of hnRNP K expression is the most in the group of metastatic lymph node.3. The expression of hnRNP K is closely correlated with the size of tumour and clinicopathological stage whlie it was negatively associated with the pathological type, the degree of differentiation of tumor cells and metastasis of the lymph node.4. The expression of hnRNP K is also correlated with metastasis of post operation but no relationship with relapse and survival time of post operation.
