Objective:To explore the influences of capsaicin on the expression of BAFF and aberrantly glycosylated IgA1of tonsillar mononuclear cells (TMCs) in IgAN patients and non-IgAN patients.Methods:22subjects, including11patients with IgAN and11patients with chronic tonsillitis and sleep apnea syndrome were enrolled in the study. Tonsillar mononuclear cells (TMCs) were isolated in IgAN and non-IgAN patients and cultured for3days with or without capsaicin. Concentrations of BAFF and IgAl in the culture supernatants and the level of IgAl O-glycosylation were determined by enzyme linked immunosorbent assay (ELISA). Gene expression of B-cell-activation factor (BAFF), core1(31,3-galactosyltransferase (C1GALT1), and core1β3GalT-specific molecular chaperone (Cosmc) in tonsillar mononuclear cells was examined by real-time PCR.Results:1. The BAFF production of TMCs without any stimulation was not different between IgAN patients and non-IgAN patients, however, the gene expression of BAFF and IgAl secretion were higher in IgAN patients. Meanwhile, the gene expression of C1GALT1and Cosmc and the level of IgAl O-glycosylation were significantly lower in patients with IgAN in comparison with those of non-IgAN.2. After coincubated with capsaicin for72h, IgA1and BAFF secretion and the level of mRNA encoding BAFF of TMCs in IgAN patients were significantly higher than that without capsaicin, and were also higher than that in non-IgAN patients. Whereas the level of IgA1 O-glycosylation and the levels of mRNA encoding C1GALT1and Cosmc of TMCs with capsaicin were significantly lower compared to that without capsaicin in IgAN patients, and lower than that in non-IgAN patients, either. While no significant difference was observed in non-IgAN patients.3. Pearson correlation analysis showed that there were significant positive correlations between gene expression of C1GALT1&Cosmc and the level of IgA1O-glycosylation (r=0.662, p=0.01; r=0.842, p=0.001).Conclusion:Capsaicin may induce IgAl secretion in patients with IgAN by activating BAFF expression, and bring to aberrant IgA1O-glycosylation by suppressing C1GALT1and Cosmc expression. As a result, limiting the consumption of hot pepper would be beneficial for patients with IgA nephropathy. The thesis comprises7figures,4tables and53references.
