Objective:To further study the cerebral protection by the combined treatment of ulinastatin and hypothermia after cardiopulmonary resuscitation in rats,we built a rat model of cardiac arrest-cardio pulmonary resuscitation successfully.Methods:We built the rat model of cardiopulmonary resuscitation by asphyxiation successfully,and divided them into five groups randomly: sham group, control group, mild hypothermia group, ulinastatin group and ulinastatin combined with mild hypothermia group. After cardiopulmonary resuscitation, the expressions of inflammatory cytokines TNF-a,IL-6and IL-8were tested by ELISA at three time points (1hour,3hours,6hours) respectively.Finally we detected the pathological morphologic change of brain tissue abtained six hours after surgery by optical microscopy.Results:(1) the expression of inflammatory cytokines in the serum: Compared with the control group, mild hypothermia group, ulinastatin group and combined group could inhibit the expression IL-6, IL-8, TNF-α in3hours significantly (P<0.05), the inhibitory effect of the combined group is more apparent. When the time was extended to6hours, there is no significant difference (P>0.05) among the combined group, mild hypothermia group and ulinastatin group.(2) Pathological morphologic change in brain tissue:each group had different degree of brain injury after cardio-pulmonary-cerebral resuscitation. The degree of brain injury in hypothermia group or ulinastatin group was lighter than in room temperature treated group, the combined group was obviously lighter than hypothermia group or ulinastatin group.Conclution:The combined application of Ulinastatin and mild hypothermia could inhibit inflammatory reaction caused by cardiopulmonary resuscitation and retard brain damage.the paper contains.6figures,4tables, and65references...
