BackgroundSkin squamous cell carcinoma(SCC) is a common cutaneous tumor. The incidenceof SCC has increased in China in recent years. This study investigates the clinicalcharacteristics of32patients with SCC so as to provide basis for the early diagnosis ofSCC. Cyclin D1is located in the11q13chromosome region and is an important cellcycle regulatory factor involved in the regulation of cell growth and proliferation.Up-regulation of cyclin D1has been confirmed in a variety of human primary tumors,which is implicated in the deregulation of the cell-cycle machinery, resulting inuncontrolled cell proliferation, tumor heterogeneity, invasion and metastasis. However,the expression of cyclin D1in arsenic-induced SCC is unknown, and the pathogenicvalue of their expression is unclear. This study is aimed to investigate expression ofcyclin D1during malignant transformation of HaCaT cells, and to assess its possiblefunction in arsenic-induced carcinogenesis.Objective1.To sum up the clinical characteristics of32patients with skin squamous cellcarcinoma.2. To investigate the expression of cyclin D1during malignanttransformation of HaCaT cells.Methods1.The clinical materials from32patients with skin squamous cell carcinoma whowere treated in our department from November,2009to April,2012were analyzed.2.Soft agar colony formation assay was used to observe the monoclonal formation, andthe expression level of cyclin D1was detected by westernblot method.Results1. Analysis of clinical materials showed:There were12cases whose lesionslocated on the face,10cases on the scalp,2cases on the opisthenar,3cases on the trunk,2cases on the forearm,1case on the leg,2cases on the genital area.The lesionsappeared as ulcerative type in18cases, cauliflower shape or papillary neoplasm in11cases, eczematoid plaque in2cases, scarlike nodule in1case. The higher morbidityappeared in those older over50years.2.Studies in vitro showed:The monoclonalformation rate of HaCaT cells in the16weeks arsenic-treated group was significantlyhigher than that in the control group by soft agar colony formation assay.The cyclin D1expression levels of HaCaT cells in the16weeks arsenic treated group weresignificantly higher than that in the control group.Conclusion1. Skin squamous cellcarcinoma occurred more at the exposure area with the most common type beingulcerative type.2. The monoclonal formation of HaCaT cells treated with arsenicincreases,and the expression level of cyclin D1significantly increases. Cyclin D1maybe related to arsenic-induced malignant transformation of HaCaT cells.The studyprovides potential novel target for future therapeutic development in skin cancer.
