Objective: This study was designed to investigate the expression of P57kip2and cyclinD1mRNA in the (OSCC) and normal oral tissues (NOT) from protein molecular level.We analyze the relationship of P57kip2and cyclin D1gene with patients’ the clinical andpathological features,such as age, sex, clinical stage, pathological grade and lymphnode metastasis, and also explore the relationship between the two genes with OSCCdevelopment, invasion and clinical features. in order to assist the clinical workeffectively.Method:This study selected42cases of surgical resection of OSCC and14cases NOTas experimental subjects,42cases of OSCC were selected as the the case group,and14cases NOT for the control group. Reverse transcription-polymerase chainreaction(RT-PCR) was used to detect the expression of p57kip2and Cyclin D1gene inthe two groups of subjects in an effort to obtaining experimental data for analysis. Allof the samples must be selected with the following criteria: the first case of a definitediagnosis, no history of other head and neck cancer, no chemoradiotherapy history andthe history of anti-tumor therapy, no other malignancies history and no family historyof cancer. All fresh samples were saved in the-80℃ultra-low temperature refrigeratorin half an hour.Results:1. The expression of p57kip2and Cyclin D1gene were detected to a in varyingdegrees in42cases of OSCC and14cases NOT. The positive rate of p57kip2mRNA inOSCC was obviously lower than that in NOT(P<0.05). The comparative content of p57kip2mRNA in OSCC was (0.540±0.160), which was much lower than the(0.902±0.057) in NOT(P<0.01). The positive rate of Cyclin D1mRNA in OSCC wasevidently higher than that in NOT(P<0.01). The comparative content of Cyclin D1mRNA in OSCC was (1.164±0.095), which was much higher than the (0.660±0.033) inNOT(P<0.01).2.The positive expression rate of p57kip2gene and CyclinD1gene in oral squamous cellcarcinoma was no relationship with patient’s age and gender, but they were correlatedwith the clinical stage, pathological grade and lymph node metastasis of tissues.3.Spearman correlation analysis: The comparative content of p57kip2and Cyclin D1mRNA in OSCC were negative correlation(r=-0.894; P<0.01).
