Objective:Use the Estrogen-progestin load method to establish the rat model of uterine fibroids,explore the mechanism of Xiaoji granules treating uterine fibroids by observing the effects of different doses of Xiaoji granule on the rat model of uterine fibroids on uterine weight,hemorheology,T lymphocyte subsets (CD3+、CD4+、CD8+、CD4+/CD8+) and TNF-α.Methods:72female SD rats were randomly divided into six groups:blank group, model group, positive control group and high,medium and low-dose groups of Xiaoji granules,all groups use the Estrogen-progestin load method to establish the rat model of uterine fibroids except the blank group.From the beginning of the ninth week,give the corresponding drugs to treat uterine fibroids.On the seventeenth week,measure the weight of rat and the weight of the uterus and calculate the coefficient of uterus, detect the indexs of hemorheology, T lymphocyte subsets and the lever of TNF-α.Use SPSS16.0to count and anlyse the results.Result:1The weight of the uterus and the coefficient of uterus of model group are higher than that of blank group,there is significant difference(P<0.01). Contrast to the model group, the coefficient of uterus of Guizhi Fuling Capsule group and high,medium-dose groups of Xiaoji granules decline, there is statistical significance(P<0.05);There is statistical significance between medium-dose group of Xiaoji granules and model group on the weight of the uterus (P<0.01).2There is statistical significance between model group and blank group on the indexs of hemorheology (P<0.01). There is statistical significance when the indexs of hemorheology of Guizhi Fuling Capsule group and high,medium-dose groups of Xiaoji granules contrasting to the indexs of hemorheology of model group (P<0.01). Contrast to the level of plasma viscosity and low shear rate of Guizhi Fuling Capsule group, high,medium-dose groups of Xiaoji granules are significant(P<0.01).3There is statistical significance between model group and blank group on the lever of T lymphocyte subsets (P<0.01). Contrast to the level of T lymphocyte subsets of model group.Guizhi Fuling Capsule group and high,medium and low-dose groups of Xiaoji granules are significant(P<0.01).There is significant difference between medium-dose group of Xiaoji granules and Guizhi Fuling Capsule group on the lever of CD8(P<0.01).4There is statistical significance between model group and blank group on the lever of TNF-α(P <0.01). Contrast to the level of TNF-α of model group,Guizhi Fuling Capsule group and high,medium and low-dose groups of Xiaoji granules are significant(P<0.01). There is significant difference between the lever of TNF-α of medium-dose group of Xiaoji granules and Guizhi Fuling Capsule group P<0.01. Conclusion:Xiaoji granules probably by improving the indexs of hemorheology of rat, promoting blood circulation, reducing local inflammatory exudation, reducing the level of TNF-a, inhibiting local inflammatory reaction, regulating the immune function,affecting the hyperplasia of cells of uterine smooth muscle of rats to achieve the effect of inhibition of tumor growth,promotion of tumor elimination.Comprehensive analysis,the effect of middle-dose group of Xiaoji granule was better than other therapy groups.
