| Objective: The purpose of this subject is to observe the effect of Si-Ni-Decoction to the inflammatory response and immune function on septic rats and to discuss its possible mechanism.Methods:Sixty-six SD rats were randomly divided into normal control group, normal saline group (NS group) and Si-Ni-Decoction group (SND group). After intraperitoneal injection of LPS (5mg/kg) to establish septic rats model, SND group received Si-Ni-Decoction with gastric infusion while NS group were given equal dose of normal saline in same way. Respectively collect serum samples at2hã€12hã€24hã€48h。72h after LPS to determine the concentration of inflammatory mediators such as IL-1ã€IL-6ã€IL-10ã€TNF-a, the expression level of HLA-DR, and observe the pathological changes of intestinal mucosa under electron microscope.Results:The concentration of IL-1, IL-6, IL-10and TNF-a in NS and SND group were significantly higher than normal group at2h after LPS, no statistical differences of IL-1ã€IL-6and IL-10in NS group compared with SND group (respectively2.47±0.14vs2.48±0.11,62.52±2.12vs62.55±1.46,86.66±6.12vs86.52±8.44, P>0.05).Meanwhile, the concentration of TNF-a has a significant statistical difference in both group(46.66±2.14vs24.46±2.54, P<0.01). After LPS12h to72h, the level of IL-1ã€IL-6ã€IL-10and TNF-a has a significant statistical difference in two group (P<0.01).The expression level of HLA-DR has no statistical differences in two groups at2h after LPS (4.86±0.15vs4.85±0.17, P>0.05),but it showed a great significant statistical difference at48h,72h (respectively2.74±0.16vs4.21±0.12,2.27±0.12vs3.80±0.09, P<0.01).The intestinal mucosal pathology showed the intestinal mucosa were inflammatory cells infiltrated obviously and fluffy was damaged severely in both NS and SND group at2h after LPS. The reduction of the inflammatory cells infiltrating in SND group was even better after12h and the intestine villi reparation was more complete compared with NS group.Conclusion:Si-Ni-Decoction has two-ways adjustment in regulating the homeostasis of pro-inflammatory and anti-inflammatory of the excessive systemic inflammatory response: it can reduce the serum pro-inflammatory factor expression, and increase anti-inflammatory factor expression. It can protect intestinal function and improve the immune status of septic rats in the way of reducing intestinal mucosal edema and inhibiting the excessive inflammatory response of intestinal mucosal, thus improved the prognosis of sepsis. |
