Objective:To study how much hypoxia-inducible factor-1α, vascular endothelialgrowth factor-a, matrix metalloproteinases-9express in the invasive and non-invasivemeningiomas, and whether those genes expression relate with the microvessel densityof meningiomas, to further investigate those expressions of HIF-1α, VEGF-A,MMP-9associated with aggressive biological behavior of meningiomas, to provide atheoretical basis of postoperative anti-angiogenesis therapy.Methods:According to the world health organization (WHO)2007the centralnervous system tumors meningioma classification and the exam of iconography andthe intraoperative findings, meningioma is divided into invasive meningiomas andnon-invasive meningiomas two groups. In this study we use the immune-histochemistry to evaluate the microvessel density (MVD) by CD34, as use thewestern blot and RT-PCR to detect the HIF-1α and VEGF-A and MMP-9expressionin meningiomas in the protein and mRNA level. At last we use statistical analyzerelationships of the HIF-1α and VEGF-A with the MVD and invasiveness ofmeningiomas.Results:The MVD in invasive meningiomas is higher than in normal groups andnon-invasive meningiomas, which has the difference(p<0.05); the expression ofHIF-1α and VEGF-A in invasive meningiomas associates with the MVD and has thesignificant difference(r1=0.930, r2=0.916, p<0.01); from the protein and mRNA level,the expression of the HIF-1α and VEGF-A and MMP-9in invasive meningiomas ismore than the expression in normal brain tissues and non-invasivemeningiomas(p<0.05), what has the statistical meaning.Conclusion:The expression of HIF-1α, VEGF-A, MMP-9can promote theangiogenesis and invasiveness of meningiomas, as those genes can be anti-angiogenesis therapy molecular targets, and MVD may reflect the invasiveness of meningiomas indirectly. The study provides a theoretical basis of postoperativemolecular targeted therapy.
