Objective:To study radiation sensitizing effect of Plumbagin on human tongue cancer cellline Tca-8113.Methods:1.The Tca-8113cells were cultured in vitro.2.MTT assay was employed to examine the growth status of the cells anddetermine the optimum dose for research.3.The flow cytometry was applied to detect the apoptosis ratio of Tca-8113cellsin each group.4.Clongenic assay was used to detect the survival of cells and confirm theradiation sensitizing effect of plumbagin.5.Expression of nuclear factor-κB was assessed by Western Blot.Results:1.The optimum dose of plumbagin on Tca-8113cells is1.42μm/L for24h.2.The flow cytometry assay indicated Plumbagin combination with X-ray induceTca-8113more apototsis than treated with Plumbagin or X-ray alone.3.The colony forming assay suggested the enhancement of radiative sensitizionin virtue of plumbagin,SER is1.28.4.Activation of nuclear factor-κB after treated by X-ray was repressed byPlumbagin.Conclusion:Plumbagin can enhance the radiosensitivity of human tongue cancer cell lineTca-8113. The repression of NF-κB might be the main mechanism of radiosensitivityenhancement.
