Objective :To study the effect of plumbagin on Epithelial-Mesenchymal Transition(EMT)and underlying mechanisms in human tongue squamous cell carcinoma cells.Methods:1.The MTT assay was applied to examine the effect of PLB on cell viability.2.Transwell assay was performed to examine the invasion capabilities.3.Scratch test was performed to examine the migration capabilities.4.The flow cytometry was applied to test the production of ROS levels.5.The expression level of EMT and p38 MAPK proteins were determined bywestern blotting.Results:1.PLB inhibited the proliferation of CAL27/CDDP cells,the value of IC50 in 12?24?48h are 10.3?M,3.1?M,1.5?M.2.PLB upregulates the expression level of E-cadherin while downregulating the expression level of Vimentin and Slug and PLB can inhibite the invasion and migration capabilities.3.After the treatment of 1?M PLB for 24 h,the cellular ROS level was boosked significantly.4.when cells was coincubated with GSH,the expression level of E-cadherin,Vimentin,Slug and the ratio of p-p38MAPK/p38 MAPK was abolished,compared to PLB-treated.5.p38 MAPK activator(Anisomycin)can ruduce PLB-induced EMT inhibition while p38 MAPK inhibitor(SB203580)can promote PLB-induced EMT inhibition.Conclusions:PLB inhibits epithelial mesenchymal transition via ROS/p38MAPK-mediated pathway in human tongue squamous cell carcinoma cells...
