Objective:To investigate the changes between protein location and expression levels of5-hydroxytryptamine2C receptor in Hippocampus,Frontal cortex,Hypothalamus andamygdale and the changes of protein activaty of PLCβ、DAG、IP3and PKC inHippocampus of Premenstrual Syndrome Model Rats with GanQi Depressionsyndrome,which gives apreliminary discussion on the pathogenesis of PMS with liver-qidepression syndrome, as well as researches the intervention mechanism Shuyu Capsule forthe disease.Methods:1After the vaginal smear examination and open field test, we choose the Wistarrats of health and female into test, chronic restraint stress preparation PMS qi depressionsyndrome rat model, then build PMS model rats with GanQi depression syndrome byMulti-factor method of Emotional factors stimulation,and evaluate all the group rats by bodyweight、adrenal coefficient、open field test and macro-behavior observation.2Then weadopt the immune fluorescence double-tagging technology to detect the protein distributionof5-HTR2C in Hippocampus,Frontal cortex,Hypothalamus and amygdale in the normalgroup rats,model group rats, Shuyu treatment group rats and Fluoxetine treatment grouprats.The expression level of5-HTR2C in Hippocampus,Frontal cortex,Hypothalamus andamygdale was evaluated by Westernblot.3detected by ELISA and Western blot in thenormal group rats, Sham group rats,model group rats, Shuyu treatment group rats, Fluoxetine treatment group rats and m-cpp gpoup rats PLCβ、DAG、IP3、PKC level inHippocampus to clarify Shu Yu capsule on the5-HTR2C-mediated downstream signalingpathway regulation.Results:1After the rats macro modeling results: Compared with normal group, modelgroup rats significantly decreased body weight (P <0.01); Compared with model group,Shu Yu Capsule significant weight group and the fluoxetine group increased (P <0.01);compared with normal group, liver depression model rats unresponsive, reduced activity,open field horizontal, vertical, total score were significantly lower (P <0.05or P <0.01);compared with the model group ratio, Shu Yu and fluoxetine group after treatmentadministration is sensitive to increased activity in the rat open field horizontal, vertical,total score were significantly increased (P <0.05or P <0.01).2Immunofluorescenceresults showed that: Compared with normal rats, liver depression model rats hippocampus,frontal area, hypothalamus, amygdala5-HTR2C receptor cells arranged sparsely cluttered,distribution has not changed, Shu Yu group and fluoxetine cerebral distribution of eachgroup are arranged without exception. Western blot results showed that: Compared withnormal group, model group hippocampus, frontal area, hypothalamus, amygdalasignificantly decreased (P <0.05or P <0.01); Compared with model group, Shu Yu groupand fluorine West statin group compared,5-HTR2C expression levels were significantlyincreased (P <0.05or P <0.01).3. ELISA results showed that: Compared with normalgroup, model group rats hippocampus DAG, IP3was significantly decreased; Comparedwith model group, agonist group, Shu Yu group and fluoxetine group DAG, IP3contentwas significantly increased (P <0.05or P <0.01). Western blot results showed that:Compared with normal rats, the PLCβ model hippocampal expression, PKC activity wassignificantly decreased (P <0.05or P <0.01); Compared with model group, agonist group,Shu Yu group and fluoxetine group PLCβ expression, PKC activity was significantlyincreased (P <0.05or P <0.01)Conclusion: Chronic restraint stress mainly through multivariate modeling methodsuccessfully copied qi depression syndrome premenstrual syndrome rat model; PMS qidepression syndrome may occur with rat hippocampus, frontal area, hypothalamus, amygdala,5-HTR2C expression and hippocampus PLCβ, DAG, IP3, PKC activitydecreased abnormal about; Shu Yu capsule can effectively correct the hippocampus,frontal area, hypothalamus, amygdala and hippocampus5-HTR2C expression in the PLCβ,DAG, IP3, PKC’s decreased activity of the state, which may be Shu Yu qi depressionsyndrome treating PMS central mechanisms.
