Objective Establish dominant negative JNK3(dnJNK3)transgenic mice,using constitutively active TAK1(caTAKl)transgenic mice and newly established the deactivation model to build caTAKl/dnJNK3 transgenic mice model.We used these transgenic animal models to study the effects of JNK3 deactivation on tooth enamel formation.To further search the downstream signaling molecules of TAK1-MAPK signaling pathways during enamel development,we study the effect of JNK3 in TAK1-MAPK signaling pathways on enamel development from histological level,thereby laying the foundation for the study of the mechanism of TAK1-MAPK signaling pathways in enamel formation.From the level of the histologic preliminary study TAK1-MAPK signaling pathways of enamel formation and the influence of the development.Methods The relative expression level of MAPK genes in TAK1 transgenic mice were detect.Immunohistochemistry detect the JNK1,JNK2,JNK3 protein expression level of five days after birth of WT mice.Western Blot detect the protein expression of JNK3 of mandibular between caTAK1 and wild-type mice.Establish dominant negative JNK3(dnJNK3)transgenic mice,the dnJNK3 mice were produced by microinjection method producd by Cyagen Biosciences Inc.Then the genotype of young mice were detected by PCR and the expression of dnJNK3 was detected by RT-PCR.Then built caTAK1/dnJNK3 transgenic mice model.The morphology of enamel were observed by SEM.Results The JNK3 expression level in caTAK1 mice mandibular significantly raised.The transgenic mice of dnJNK3 gene was established.The expression of deactivation JNK3 gene in deactivation JNK3 mice jaw was specific.The dental enamel morphology between caTAKl/dnJNK3 transgenic mice and wild-type mice showed no significant difference in the first month after birth.Conclusion The JNK3 gene expression level in caTAK1 mice mandibular significantly raised.The enamel of the caTAKl/dnJNK3 transgene mice is similar with WT mice.JNK3 maybe is the downstream signaling molecule of TAK1-MAPK signaling pathway during enamel development.
