Objective: To study the inhibitory effect of matrine on cell line TF-1inerythroleukemia, and the role in the expression of SALL4and downstreamWnt/β-catenin signaling pathway.Methods: The inhibitory action of matrine on the proliferation andapoptosis in TF-1cells was detected with MTT assay.Apoptosis wasexamined by flow cytometry. The expression levels of SALL4mRNA andthe components of Wnt/β-catenin signaling pathway(β-catenin, C-myc,Cyclin DI)were tested by RT-PCR. The expression levels of SALL4proteinwas measured by Western blot.Results:1.The proliferation of TF-1cells were inhibited bymatrine(P<0.01),dependent of time (48h IC501.1413g/L,72h IC500.7998g/L).2.Matrine inhibited the growth and survival rate of TF-1cells inconcentration-dependent manner (41.15±11.75)%,(60.15±4.84)%treatedby1,0g/L and2.0g/L respectively),which was statistically different from the control groop(19.06±1.41)%.3.It showed in RT-PCR that the expression levels of SALL4mRNA andthe components of Wnt/β-catenin signaling pathway(β-catenin, C-myc,Cyclin DI) significantly decreased (P<0.05)after intervened by matrine.4.Moreover, dependency was observed through analysis on theexpression of SALL4and β-catenin, C-myc, Cyclin DI,(rs=0.912,P<0.01;rs=0.818,P<0.01; rs=0.832,P<0.01).4.Western blot revealed the expression levels of SALL4protein weredecreased (P<0.05).Conclusion: Matrine probably play a role in inhibiting the proliferationand inducing apoptosis through inhibiting the expression of SALL4geneand Wnt/β-catenin signaling pathway.
