Objective :Matrine (Mat) induced human erythroleukemia TF-1 cell apoptosis and effect on SALL4 expression.Methods:Different concentrations of the Mat (0.5,1.0,1.5,2.0 g/L) were cultured in vitro in TF-1 cells at different time (24,48,72h); TEM morphology of TF-1;MTT assay of cell proliferation; flow cytometry determination of the cell cycle; Annexin V and PI double staining method to detect cell apoptosis; RT-PCR detection SALL4 mRNA expression.Results: 1. Mat (0.5-2.0g / L) after 24,48,72h the role of TF-1 cells were inhibited the proliferation (P<0.01), in the concentration range of inhibition in a dose and time dependent, 48h half inhibitory concentration ( IC50) was 1.0g/L.2. Mat role of TF-1 cells after 48h: Compared with the control group, the proportion of G0/G1 phase cells increased, S phase cells decreased (P<0.01).3. Flow cytometry to detect apoptosis were 8.6%, 11.21%, 15.26%, 17.63%, with the control group (5.05%) compared to a statistically significant difference (P <0.01). 4. RT-PCR results showed SALL4 mRNA expression andβ-actin (internal reference) ratio of expression, with Mat dose increased significantly decreased (P<0.05).Conclusion: Mat in a certain range of concentration and time on the TF-1 cells proliferation, The mechanism is to make the cells G0/G1 phase arrest; Mat can inhibit SALL4 gene expression and induce cell apoptosis.
