Objective To investigate the effects of Zhaqi jiangtang Decoctionon the expression of TGF-β1、VEGF and nephrin in kidney of rats withdiabetic nephropathy,and the possible mechanisms of its renal protection.Methods60SD rats,8weeks old,200±20g, a week’s adaptivefeeding.Then the rats were randomly selected8as the normal group(N,n=8), the rest of them injected STZ to establish the model rats of diabetic.After4weeks, the model rats of DN were divided by random numbertable into5groups: Model group(M,n=10), Glibenclamide group(G,n=10), Zhaqi jiangtang Decoction high-dose group(ZG, n=10), Zhaqijiangtang Decoction mid-dose group (ZZ, n=10), Zhaqi jiangtangDecoction low-dose group(ZD, n=10). Then the relevant drugs weregiven intragastrically to different groups respectively. At the end of10thweek, take Inner canthus blood to measured BUN, Scr by routineanalyse methods; take renal tissues to detect the pathological morphologychanges and ultrastructural changes; the expression of TGF-β1was detected by immunohistochemical staining; the expression of VEGF wasdetected by immunofluorescence staining; the expression of nephrinmRNA and nephrin protein were determined by applying RT-PCR andWestern blot.Results In model group, BUN, Scr in blood were higher than thenormal group(P<0.05), renal tissue pathological morphology changesand ultrastructural changes were significant, the expression of TGF-β,VEGF was significantly higher than the Normal group(P﹤0.01), and theexpression of nephrin mRNA and protein was significantly lower than theNormal group(P﹤0.01). Compare with model group, other groupsimproved significantly, especially in Zhaqi jiangtang Decoction high-dose group and Glibenclamide group (P <0.05), in which the blood BUN,Scr were decreased, the expression of TGF-β1, VEGF weredown-regulated, the expression of nephrin mRNA and nephrin proteinwere increased.Conclusion Zhaqi jiangtang Decoction has a protective effect onthe kidney of rats with diabetic nephropathy, and the mechanisms may bethrough down-regulation the expression of TGF-β1, VEGF, up-regulatethe expression of nephrin mRNA and nephrin protein to reducegeneration and precipitation of glomerular mesangial matrix, andreduce podocyte injury.
