Abstract/Summary:
Objective:To investigate the phaenotype character of specific memory T-cell long term aftermeasles vaccine inoculation on child and provide theory direction for long time effectand new inoculation strategy after measles vaccine inoculation.Methods:(1) To choice32children of3-10years old inoculated measles vaccine on basis ofthe head of a family in longgang Maternal and Children Health hospital inShenzhen between2003.12-2010.01.(2) To collect3ml peripheral vein blood and isolate and cultivate theleukomonocyte added to measles vaccine. control group(non-add to measlesvaccine) was put in5%CO2incubator and had been cultivated for50hours in37℃.To detect the cell surface molecule CD4/CD8、CD45RA、CD45RO、CD62L、CCR7with FCM.(3) Experiment group was put in5%CO2incubator and had been cultivated for50hours in37℃and continued1hour cultivation after added in CCK-8.To detectcell multiplication with CCK-8and calculate the index of cell multiplication.Reslts:(1) The amount of CD4+T cell was more than CD8+T cell in the peripheral bloodperipheral blood monouclear cells(PBMC) by the approach of FCM detectingCD4+and CD8+T cell in experiment group, there was significant difference(P<0.05).(2) By the approach of the FCM detecting CD4+/CD8+CD45RA+naive T cellsubpopulation, the amount of CD4+/CD8+CD45RA+naive T cell were all above70%in control group and experiment group; the amount of CD8+CD45RA+ naive T cell were all above80%, there was not significant difference(P>0.05).At the same time three group were divided and compared according to antibodylevel, and the difference of three groups was not significant(P>0.05).(3) By the approach of the FCM detecting CD4+/CD8+CD45RO+naive T cellsubpopulation, control group reached to28.12%, experiment group29.55%,there was not significant difference(P>0.05).; the amount of CD8+CD45RO+incontrol group reached to17.35%, experiment group21.89%, there was notsignificant difference(P>0.05).; At the same time three group were divided andcompared according to antibody level, and the difference of three groups wasnot significant(P>0.05).(4) According to the express of homing receptor(CD62L)and chemokine receptor(CCR7)of CD4+T memory cell, the amount of CCR7+subpopulation T cell incontrol group reached to84.37%, experiment group84.47%; CD62L+subpopulation T cell in control group reached to93.60%, experiment group95.00%, there was not significant difference(P>0.05).; the amount of CCR7+subpopulation CD8+T cell in control group reached to47.24%, experimentgroup50.55%; CD62L+subpopulation T cell in control group reached to61.30%, experiment group65.89%, there was not significant difference(P>0.05).(5)To analyze the CD45RA+、CD45RO+、CCR7+and CD62L+subpopulation of theCD4/CD8cell according to the different year (3-5,6-7,8-10) after vaccineinoculation, there wer not significant difference(P>0.05).(6)To detect specific cell multiplication after vaccine stimulus, the result showed thatthe cell multiplication index all exceed0.8,17examples exceed1.0and reachedto54.84%. Conclusion:(1) Masern virus infection is intra-cellular infection,so the significance of theresearch on memory cell through measles vaccine inducing organism immunityis very important. The research indicated that CD4+T cell was the ascendant cellin T cell after measles vaccine inoculation to child, the amount of naiveCD45RA+T cell was above70%-80%.(2) According to the express of the CD45RA-/CD45RO+, the amount of the CD8+memory T cell reached to17%, CD4+memory T cell reached to28%-29%. Theamount of memory T cell was not related to antibody level, and the CD4+memory T cell was primary phaenotype character.(3) The research according to the express of CCR and CD62L further discovered thatmemory T cell subpopulation possessed the character of multiplicity and thecentral type memory cell was primary phaenotype character; The amount of cell(cell multiplication exceeding1) reached to54.84%, that showed specificmemory cell proliferate well.
