Objective: To detect the children’s serum antibody and cytokines secreted bydifferent T cell subsets after vaccination with measles vaccine, to evaluate itspersistence of humoral immunity and cellular immunity, so that we can providetheoretical basis for measles vaccine immunization strategies.Methods:32children who have vaccinated with measles vaccine for3~10yearswere chosen as healthy volunteers. Every volunteer takes5ml peripheral blood,2mlwas used to detect measles antibody with ELISA after separation of serum,3ml wasused to separate lymphocytes. Lymphocytes separated as measles vaccine group andcontrol group were cultured50hours under the condition of37℃,5%CO2, afteradjusting cell number to1×106/ml with10%FBS RPMI-1640. Then we take thesupernatant to detect TNF-α and IL-10with Cytometric Bead Array(CBA) and IFN-γ,TNF-α, IL-4and IL-10with ELISA.Results: The mean of32cases’ antibody concentration is115.03IU/L and thestandard deviation is95.20IU/L. There are great differences between the antibodylevels. The antibody levels of high response group were obviously higher than theother two groups (p<0.05). TNF-α and IL-10were detected with CBA and the medianof the group with measles vaccine stimulation is2021.81pg/ml and2566.93pg/ml.The median of the group without vaccine stimulation is1481.72pg/ml and1861.89pg/ml. There is statistical difference (p<0.05) for TNF-α, there is statistical difference(p<0.05) for high response group, response group and low response group. There isstatistical difference (p<0.05) for IL-10, there is statistical difference (p<0.05) forresponse group and low response group, there is no difference for high response group.For IFN-γ, TNF-α, IL-4and IL-10detected with EIISA, the mean of the group withmeasles vaccine stimulation is152.07pg/ml,160.92pg/ml,182.55pg/ml and554.46pg/ml, the mean of the group without vaccine stimulation is187.31pg/ml,129.57pg/ml,190.87pg/ml and398.94pg/ml. There is statistical difference (p<0.05) forTNF-α and IL-10, there is no difference for IFN-γ and IL-4.Conclusion: There is a high positive rate of antibody of the children with measles vaccination for3~10years (81.2%). The mean of antibody levels is115.03IU/L. Butthere is no obvious relationship between the antibody levels and age, gender,vaccination times or the inoculation time. TNF-α, IL-10increased significantly in thechildren who had been vaccinated against measles3~10years ago when they werestimulated with measles vaccine again. It suggests that there are memory T cells andthey can lead to rapid response when exposure to antigens.
