Objective:①Invesigate inhibiting effects of Interleukiu-24proteins combined with Cisplatinon SKOV3and SKOV3/ddp of human ovarian cancer cell growth in vitro;②Invesigate the effects of Interleukiu-24proteins combined with Cisplatin inducingapoptosis,the chang of cell-cycle and apoptosis-related genes on SKOV3andSKOV3/ddp of human ovarian cancer cell in vitro;③Invesigate inhibiting effects of Interleukiu-24genes combined with Cisplatin onSKOV3of human ovarian cancer cell in vitro;Methods:①The SKOV3and SKOV3/ddp cells were infected with rhIL-24alone,DDPalone,and rhIL-24combined with DDP, the SKOV3cells were infected withAd-hIL-24alone,DDP alone and combined with Ad-hIL-24and DDP to inhibitedthe growth of ovarian cancer cell;②The growth-suppressing of SKOV3and SKOV3/ddp were assessed by CCK8assay, then count the suppression ratio;③The changes in apoptosis-inducing and cell-cycle of SKOV3and SKOV3/ddpwere detected by flow cytometry;④The apoptosis-related genes alteration of SKOV3and SKOV3/ddp were assessedby GeXP genetic analysis system;Results:①SKOV3and SKOV3/ddp cells enter the exponential phase in the third day underthe right conditions;②rhIL-24can suppressed SKOV3and SKOV3/ddp cells and no toxicity to normalcells,the two cells suppression ratio is30.7%and21.8%respectively, the suppression ratio of group of rhIL-24combined with Cisplatin is35.3%and32.6%respectively, It showed a significant difference compared with the rhIL-24alone and DDP alone (P<0.05), rhIL-24can enhances the effect ofchemotherapy.③The apoptotic rates detected by flow cytometry in rhIL-24alone group,DDPalone group and combined group showed a significant difference compared withthe negative control group.Furthermore,cell cycle detection indicated that thepercentage of SKOV3in G2phase increased in rhIL-24alone group. Thepercentage of SKOV3/ddp in G2and S phase increased in rhIL-24alone group.④The genes expression level of Bax, caspase-3, Rb was obviously unregulated;and Bcl-2, survivin was down regulated in SKOV3and SKOV3/ddp cells afterinduced by rhIL-24. All results were more obviously when all genes mentionedabove induced by rhIL-24combined with DDP.⑤Ad-hIL-24can suppressed the growth of SKOV3cells of human ovariancancer.The combined group showed a significant difference compared with theAd-hIL-24alone and DDP alone (P<0.05),Ad-hIL-24can enhances the effect ofchemotherapy.Conclusion:①Interleukiu-24proteins and genes can suppressed the growth of human ovariancancer.of SKOV3and SKOV3/ddp cells in Vitro, Furthermore,combined groupshowed a significant difference compared with alone group, both the rhIL-24andAd-hIL-24and can enhances the effect of chemotherapy. Otherwise can induceapoptosis human ovarian cancer of SKOV3and SKOV3/ddp cells;②Interleukiu-24proteins can induce the change of cell-cycle alteration humanovarian cancer of SKOV3and SKOV3/ddp cells; meanwhile,can change itsapoptosis-related genes.
