| Objective: This study was to investigate those targets relevant to Aβ metabolism inHippocampus tissues of rat Alzheimer’s disease(AD) model treated by Traditional ChineseMedicine(TCM) HeiXiaoYao powder with the method of screening to provide experimentalbasis and scientific evidence.Methods: Half of male and female wistar rats ware chosen to establish AD model byinjecting Aβ25-35amyloid into hippocampal of rats,and all rats were divided into6groups,which were sham operation group,model group,positive control group and Hei Xiaoyaopowder treatment groups of different dosages. Among them, rats of sham operation groupwere treated with same volume saline injecting in hippocampal, rats of other groups weretreated with same volume saline injecting Aβ25-35amyloid solution in hippocampal tissues.After7days of AD model-making operation, rats of HeiXiaoYao powder treatment groupsand positive control group were administrted with extraction solution of HeiXiaoYao powderand Huperzine solution by gavage administration for28days (3ml/kg/day)respectively, andrats of sham operation group and model group were administrated with saline gavageadministration of same volume and same times. After completed treatment, rats of all groupswere killed and dissected to obtain hippocampal tissue for RNA isolation and tissue proteinsextraction.(1) Rat gene Microarray was employed to screen gene expression profiles ofhippocampal tissue in AD model rats,and qRT-PCR was used to validate a few aberrantlyexpressed genes;(2) PCR-Array was employed to screen aberrantly expressed genes ofhippocampal tissues between sham operation group and model group, and qRT-PCR was usedto validate a few aberrantly expressed genes; Western blotting method was used to validatethose aberrantly expressed targets changed by HeiXiaoYao powderResults:(1) Black Xiaoyao podwer regulate the expression of Zfp37,Znf483,ZIC4about zincfinger protein to affects occur of AD.(2) Black Xiaoyao podwer regulate the expression of Wisp-1, Crebbp,Igfbp-1,Casq2about wnt signaling pathway to affect Aβ metabolism and Tau protein abnormallyphosphorylated;(3) Black Xiaoyao podwer regulate the expression of Clu,Map-2,AChE,Apba1toaffect deposition and removal of Aβ amyloid. Conclusion: The roles and mechanisms of HeiXiaoYao powder maybe related to theinhibition of A β amyloid deposition and eliminate in hippocampal tissues.beside this, its rolesand mechanisms of HeiXiaoYao San powder treatment on AD model rats maybe related to itsability to regulate and protect multiple neurons, and to affect the targets involved in nerveregeneration pathways. |
