Background:Bushen Yizhi Formula (BSYZF) is established by professor Lai, which mainly composed of common cnidium fruit, medlar, ginseng, radix polygoni multiflori, cortex moutan, borneol and other kinds of herbs. Previous studies of my department showed that BSYZF can improve the AD animal models of learning and memory behavior and the condition of pathological tissue damage. Based on this, we conducted a study about the influence of Bushen Yizhi Formula (BSYZF) on Aβ25-35 hippocampal injection memory impairment model.Objective:Applying intrahippocampal injection of Aβ 25-35 to establish murine models of Alzheimers disease to study the effects of Bushen Yizhi Formula (BSYZF)on behavior of the model mouse.Methods:Kunming (KM) mice randomly divided to one of six groups:Sham group, Model group, Aricept group, Low dose group, Middle dose group, High dose group,3 days after Aβ 25-35 injection each group was administered with saline or BSYZF by oral gavage continuously for four weeks.The Morris water maze(MWM) task, object recognition task (ORT) was used to evaluate the learning and memory functions in mice. Mice were sacrificed and cerebral coronal slices were used for HE staining,Nissl’s staining.Results:(1) the behavioral testsMorris water maze results:compared with normal group, model group mice in the navigation test escape latency prolonged (P<0.05), through the original table number in space exploration tests were significantly reduced (P<0.05), in the retention period of the original platform quadrant significantly reduced (P<0.05); Compared with model group, high dose group and the escape latency navigation tests positive medicine group were significantly reduced (P<0.05), through the original table number in space exploration tests were significantly increased (P<0.05), high dose group is in the retention period of the original platform quadrant significantly increased (P<0.05).New method of identification:recognition test of new things, be familiar with the differences between each position preference index in mice during without statistical significance; Test phase of the control group and treatment group to new things in mice preference index is higher than the model group, but there was no statistically significant difference.(2) the pathological dyeingCompared with normal group, model group hippocampal pyramidal cells with the number of neurons decreased, cell shrinkage, nuclear pyknosis of triangle or polygon, nucleus and cytoplasm are hyperchromatic concentrate. Neuron apoptosis loosely arranged, disorder, depigmentation phenomenon is obvious, level and not clear, less often gather in groups, some occupy hippocampal pyramidal cells with a completely.Conclusion:1, the water maze test results suggest Bushen Yizhi Formula (BSYZF) can to A certain extent, improve Aβ 25-35 hippocampal injection to memory impairment model of learning and memory ability in mice.2, new recognition experiment, familiar with the differences between each position preference index in mice during without statistical significance; measuringTest phase of the control group and treatment group to new things in mice preference index is higher than the model group, but there was no statistical difference meaning.3, Bushen Yizhi’Formula (BSYZF) can by improving the loss of hippocampal neurons and exceptions, cure A β 25-35 hippocampal injection to memory impairment model of learning and memory ability in mice, the AD has A certain therapeutic effect.Extracted "from the experimental results can be found in the kidney in educational party site treatment for alzheimer’s disease have a certain role, deserves further research.
