Effect Of Heixiaoyao Powder On Cognitive Memory Capacity And Proteomics Of AD Model Rats

Objective: To study the affect of Heixiaoyao Powder onAβ25-35-induced rat model ofAD-related behavior and proteomics to explore the mechanism of action ofHeixiaoyao Powder AD prevention.Methods: The Wistar rats(n=120)，half of them were female and the others were male,wererandomly divided into sham surgery rats(n=20)group and model group(n=100),the number offemale ones was the same with the male ones in each group,7days later, all rats of modelgroup received water maze tests. These eligible rats were randomly divided5groups,modelgroup,positive control group,Heixiaoyao Powder high,medium and low dose groups,20rats ineach group.Aβ25-35fragments was injected into the hippocampus to damage brain neuron tomake the rat’sAD mode1.All rats of Heixiaoyao Powder group were orally administeredHeixiaoyao Powder at low,medium and high doses(4.2,8.50,17.0g·kg/d),the positive controlgroup was orally given Huperzine Asolution0.02mg·kg/d,model group and sham groupreceived an equal volume double distilled water daily for28d. Morris water maze wasapplied to observe the behavior changes ofAD rats; we expect to learn more about the effectof Heixiaoyao Powder on proteomics of AD model rats of hippocampus by protein chiptechnology.Results:(1)This subject of study results showed that making the rat’s AD mode1by injectingAβ25-35fragments into the hippocampus of rats is a success,the model group was significantlylonger than the latency of rats in sham group(p <0.05),the difference was statisticallysignificant;(2)Morris water maze test showed that electro-acupuncture can improve the ADmodel rats in learning and memory ability,The incubation period for each intervention groupwas significantly lower than the untreated group(p<0.05),the difference was statisticallysignificant;(3)Immunohistochemical test results showed that Heixiaoyao Powder candecrease IL-1beta, TNF alpha, IL-6expression level in the serum of AD rats(p <0.05);(4)We choose the PS2,Anxa4, TGF-beta, Camk2d these four indicators by means of theproteomics studies, Heixiaoyao Powder can decrease the expression of PS2,Anxa4, TGF-beta, increase Camk2d. Conclusion:Heixiaoyao Powder can significantly improve Aβ25-35-induced rat model of ADlearning and memory,reduce cell in flammatory response and reduce apoptosis.
