Purpose: To explore the application of anesthetics’(chloral hydrate) influence on the tauphosphorylation in rat cerebrospinal fluid and hippocampus,and the correlation betweenthem.Methods: Animals:3-months old male SD rats Anesthesia group:Intraperitoneal injectionof rats with chloral hydrate500mg/kg, and to take the cerebrospinal fluid, hippocampus at0min,30min,60min,90min,120min,24h after anesthesia. Control group: Intraperitonealinjection of rats with Saline450mg/kg, and to take hippocampus after5min. Using Westernblot to detect the levels of p-tau205, p-tau212and t-tau in hippocampus, with ELISA todetect p-tau205in cerebrospinal fluid.Results: The phosphorylation tau levels in the hippocampus had an upward trendimmediately after the anesthesia, and at30-120min the levels increased significantlycompared with the control group (p<0.01). They declinced since90minutes approximatelyafter anesthesia, and approached to normal levels at24hours after anesthesia. The level ofthe phospho-tau increases over time in cerebrospinal fluid, at60-90min the level increasedsignificantly compared with the level at0min (p<0.01). The changes of the phospho-taulevel in cerebrospinal fluid and hippocampus are positively correlated (R≈0.68). It showedno significant change of total tau level in hippocampus over time.Conclusion: With anesthetic agent (chloral hydrate) acting on rat, the phospho-tau levels will increase in cerebrospinal fluid and hippocampus. The phospho-tau levels have acorrelation between the cerebrospinal fluid and hippocampus.
