Objective:By detecting the activity of NF-κB and the caspase-8,3activity, the apoptosis of podocyte under different condition and the expression of Fas to investigate the mechanism of podocytes apoptosis induced by ADR and the celluar protection of FK506.Methods and Materials:conditionally immortalized mouse podocytes were cultured in vitro, ADR was used to induce apoptosis with or without FK506.Apoptosis was measured by MTT and fluorescence-activated cell sorting (FACS).ELISA was used to meaure the activity of NF-B. The spectrophotometry was used to detect the activity of caspase8,3. To establish a role for Fas, Fas levels was measured by westernblotingResults:FK506protects podocytes against ADR induced apoptosis in podocytes. Further, the activity of NF-κB, caspase8,3and the expression of Fas in control group was significantly lower than ADR model group and the intervention group (P<0.05), the activity of NF-κB, caspase8,3and the expression of Fas in intervention group was lower significantly than ADR model group (P<0.05).There was no difference between the control group and the FK506group(P>0.05).Conclusion:FK506is prosurvival in injured podocytes. FK506can inhibit the NF-κB’activity and down-regulate the expression of Fas and the activity of caspase8, 3in ADR-induced podocytes. For the reason that, FK506prevents podocytes apoptosis from ADR induced podocytes.
