Objective To investigate the expression of Desmin and TGF-(31in adriamycin(ADM) induced podocyte injury and assess1,25-dihydroxyvitamin D3on podocyte injury model induced by ADM.Method Firstly in vitro podocyte cell were treated concentration gradient of ADM. MTT and Annexin Ⅴ-PI flow cytometry assay assessed the podocyte cell viablity and apoptosis, then real time-qPCR and immunofluorescence assay was used to detect the Desmin and TGF-(31expression. A in vitro podocyte injury model induced by a appropriate concentration ADM was established. Lastly, the effects of1,25-dihydroxyvitamin D3on this model was assessed.Results①ADM damaged the in vitro podocyte obviously. Cell viability was decreased to (82±5)%(P<0.05) when podocyte were treated for24h by1μmol/L ADM in MTT assay. With the increase of ADM concentration, the podocyte cell viability decline gradually. The podocyte apoptotic and necrosis rate was increased in a dose-dependent manner after ADM treatment. The percentage of early apopotic cell is (11±2)%with24h treatment of ADM compared to (1.3±0.3)%with no treatment (P<0.05).②In ADM injured podocyte, Desmin and TGF-(31mRNA expression was increased in a dose-dependent manner. Compared with uninjured podocyte, Desmin and TGF-(31mRNA expression were significantly higher in podocyte treated for24h by1μmol/L ADM (P<0.05).③1μmol/L ADM treated to the in intro podocyte for24h as a podocyte injury model,1,25-dihydroxyvitamin D3suppressed Desmin and TGF-β1mRNA expression in this model compared with control group (P<0.05). Desmin and TGF-β1protein was detected by immunofluorescence. The distribution of Desmin protein changed from cytoplasm to nucleus when podocyte injured.Conclusion①The ADM induced in virto podocyte injury model can use to research for future.②1,25-dihydroxyvitamin D3can significantly suppress the express of Desmin and TGF-(31in ADM induced podocyte injury, and the protect effects of1,25-dihydroxyvitamin D3in podocyte may partly contribute to it.
