Objective: To investigate the effect of IL-17 gene silencing on expression of Th17 cells and related cytokines IL-17,Treg cells and related transcription factor Foxp3 in serum as well as characteristic protein Nephrin and Podocalyxin on renal podocyte.Methods: The shRNA expression plasmid of IL-17-shRNA was constructed and transfected to adriamycin nephrosis rats model by applying letivirus mediated transfection as transfected group,While adopted the rats untreat as blank and those transfected with vector as negative group,At what the well-transfected rats were screened out by GFP,Then performed CBB to check albuminuria,Further,The numbers of Th17 and Treg cells in serum were analyzed by flow cytometry,The related level of IL-17 and Foxp3 were determined by real time quantitative PCR(qPCR),The level of Nephrin and Podocalyxin on podocyte were determined by qPCR,Western blot and immunohistochemistry respectively,Following,The ultrastructure changes of glomerular were visualized by electron microscope.Results:(1)Compared with control group,The albuminuria,Th17 cells and IL-17 were increased(P<0.05)while Treg cells and Foxp3 were reduced obviously as well as the expression of Nephrin and Podocalyxin(P<0.05),Furthermore,The increase proliferation of mesangial cells,thickness of glomerular basement,dense deposits,foot process fusion were remarkably detected through electron microscope in model group.(2)Compared with model and negative group,The albuminuria,Th17 cells and IL-17 were decreased(P<0.05)while Treg cells and Foxp3 were raised obviously as well as the expression of Nephrin and Podocalyxin(P<0.05),Further more,The mild mesangial matrix hyperplasia,intact glomerular basement,merely dense deposits,partial foot process fusion were observed apparent through electron microscope in transfetion group.T test results between model and negative group have no statistically significant difference(P>0.05).Conclusion: The abnormal activation of IL-17 involve in podocyte injury that led to massive proteinuria which caused renal injury and chronic progress in PNS,The effect of silencing IL-17 gene in adriamycin nephrosis rats show a great positive influence,What it may be expected as one of a new strategy to treat podocyte injury in primary nephrotic syndrome in future.
