Objective To study the effect of4-Hydroxyisoleucine (4-HIL) which was the activeingredient of fenugreek on insulin resistance of3T3-L1adipocyte induced by highglucose and insulin, and to investigate its possible molecular mechanism.Methods3T3-L1Adipocyte was treated with25mmol·L-1glucose with0.6nmol·L-1insulin to induce insulin resistance.4-HIL was used for the treatment andpalmitic acid for the control group.[3H]-D-glucose uptaking method was used toobserve the transporting rate. PCR was used for testing the gene expression of celluarTNF-α, and Enzyme-Linked Immuno Sorbent Assay (ELISA) was set to test thecontent of TNF-α in culture supernate.RESULTS After the treatment with25mmol·L-1glucose with0.6nmol·L-1insulinfor18h, the insulin-stimulated glucose transportation in3T3-L1adipocyte wasinhibited by63%, and the gene expression of cellular TNF-α as well as the content ofTNF-α in culture supernate increased significantly.And the palmitic acid groupshowed the similar results to the model group. Meanwhile, these were reserved byprior treatment of the cells with4-HIL.Conclusion4-HIL could significantly improve the insulin resistance of3T3-L1adipocyte induced by high glucose and insulin, which may be caused by the decline ofsecretion of TNF-α induced by4-HIL.
