| Materia Medica of all previous dynasties have performed detailed records of its efficacy,Fenugreek in invigorating kidney and strengthening yang,falling blood sugar,etc,have very good medicinal value.Much attention has been paid to galactomannan,saponins and flavonoids as the hypoglycemic ingredients of the highest levels,the separation and purification,the compatibility mechanism of the three components have been studied in depth,but the preparation is complicated,time-consuming,labor-intensive.In order to simplify the process and save the cost,this article chooses extract of Fenugreek as the research object to carry out the experiment.Firstly,orthogonal experiment was used to determine the best ethanol extraction process;secondly,definited the difference of the extraction yield of active ingredients and the chemical compositions of the extracted components between the ethanol extract and the water extract,and further study on the efficacy in the vivo and vitro of the extracted components from two kinds of extraction process,and the final extraction process was determined based on the results of the efficacy,Finally,the preparation and quality standards of capsules were studied.The research content is mainly divided into the following four aspects:1.Studies on extraction technology of hypoglycemic components from FenugreekThe ethanol extraction craft was optimized by orthogonal test with The extraction yield of flavonoids and saponins as an index,the optimal ethanol extraction process was determined as follows: solid-to-liquid ratio is 1:10;ethanol concentration is 70 %;extraction time is 2 h,3 times.By comparing the extraction yield of the active ingredients between the ethanol extract and the water extract,learned that the extraction yield of flavonoids,saponins and polysaccharides in the water extraction process respectively were 0.49%,7.18% and 10.61%;the extraction yield of flavonoids and saponins in the optimal ethanol extraction process respectively were 1.06 % and 8.24 %,The total flavonoids,total saponins,total sugar and polysaccharide yield by water extraction respectively were 0.49 %,7.18 %,22.28 % and 10.61 %.2.Analysis of the ingredients of Fenugreek extractThe UPLC-Q-TOF-MS was used to compare the aqueous extractss with the ethanol extracts in analyzing the principal components.The results showed: the total ion flow diagrams of extract ingredients respectively extracted by ethanol extraction and water are similar,and the main components are the same.a total of 36 compounds were determined,including 12 flavonoids,24 saponins.3.Studies on auxiliary hypoglycemic activity of components from FenugreekFurther study on hypoglycemic activity of components by water extraction and ethanol extraction.The results showed that components from water extract and ethanol extract all have α-glycosidase enzyme inhibition activity,its IC50 respectively were 5.83,7.27 mg/mL,α-glycosidase enzyme inhibition activity of components extracted by water extraction is stronger;in HepG2 insulin resistance experiment,the sugar consumption of cells is 2.32 mM(group of components extracted by water extraction),the sugar consumption of cells is 2.23 mM(group of components extracted by ethanol extraction),components from water extraction and ethanol extraction have similar ability of improving insulin resistance;in the diabetic mice model induced by ALX,both water and ethanol extracts have improved survival of experimental animals,reduced fasting blood glucose levels,increased glucose tolerance,elevated serum insulin levels,and promoted the synthesis of hepatic glycogen and muscle glucose,have the effect and show different activity on a visit to these different indicators.Combined with economic feasibility of the process,and ultimately determine the aqueous extract process for the study of capsule.4.Research for forming process of auxiliary hypoglycemic capsules of components from Fenugreek and content determinationThe experiment selected wet granulation to prepare content particles of capsule,based on the minimum dosage and types of excipients,hardness and formability of soft material,appearance of grain to determine the final granulating method: according to the ratio 1:1.5 of the concrete: accessories add accessories(starch)to the concentrated extract(density between 1.30-1.40)for soft material,grains granulated by 14 mesh,dried about 2 h at 65 °C,sized.The results of content determination showed that the content of the flavonoids,saponin,total sugar and polysaccharide from grains respectively were 0.16 %,2.48 %,6.47 % and 1.78 %.To sum up,this study finally selected extract by water extraction from fenugreek as the research object,maded clear its pharmacological activities in hypoglycemic,and water extraction process is simple,save raw materials,the feasibility is strong,provided theoretical basis for development and utilization of hypoglycemic health products of extract from fenugreek. |
