ObjectiveTo explore the poison effects of ephedrine on liver and protective effects of thetraditional Chinese medicine compound on pregnant and filial mice of ephedrinedamage.The behavior、 body weight、liver weight and total antioxidant capacity andglutathione S-transferase and maleic dialdehyde content and histological structure andpositive expression of c-Fos protein and TGF-β1in liver of were measured afterdifferent doses of intraperitoneal injection of ephedrine as well as intragastricadministration of the traditional Chinese medicine compound pregnant mice.75pregnant mice at the fifth day of pregnancy began to be continuouslyintraperitoneal injection with different doses (2.0g/L,4.0g/L and6.0g/L) ofephedrine (0.2mL) in the experimental groups and the same amount of saline in thecontrol group in the morning and afternoon until the growth of filial mice to15dayand Chinese medicine group injected ephedrine after1hour respectively intragastricadministration with0.2mL, the control group injected of same amount ofsaline.Changes of body weight and liver weight were detected and the activity ofT-AOC、GST and MDAcontent were respectively detected by colorimetry in the liverof pregnant mice and filial mice at each developmental period from all experimentalgroups. In addition,the changes of the liver histological structure by opticalmicroscope.The expression of c-Fos protein and TGF-β1were measured byimmunohistochemistry combined with stereology in liver of pregnant mice and filialmice at different developmental stages.Results1.Ephedrine and the traditional Chinese medicine compound affected onbehavior、liver weight and body weight of pregnant and filial mice. Ephedrine groupsdemonstrated symptoms such as running, jumping,reducing food and drink, fightingeach other,vertical tail and tremble of pregnant mice. And the liver weight and bodyweight of pregnant mice and filial mice of the ephedrine groups was lower than thecontrol groups at the different developmental period(P <0.05or P <0.01).The6.0g/L ephedrine group at25day appeared falling off hair and stillborn foetus mice. Theliver weight and body weight of pregnant mice and filial mice of the traditionalChinese medicine compound group was higher than the6.0g/L ephedrine group at thedifferent developmental periods (P <0.05or P <0.01).2.Ephedrine and the traditional Chinese medicine compound affected the antioxidant enzymes activity in liver of pregnant and filial mice,there was a greatinhibition for the T-AOC,GST activity of ephedrine groups (P <0.05or P <0.01).Thegreater the doses of ephedrine, the stronger the inhibition. But the MDA contentevidently increased (P <0.05) and was higher with the increased doses of ephedrine.Compared with the6.0g/L ephedrine group, T-AOC,GST activity of the traditionalChinese medicine compound group increased (P <0.05),the MDA content of thetraditional Chinese herbal medicine in liver evidently was lower than the controlgroups (P <0.05or P <0.01).3.Ephedrine and the traditional Chinese medicine compound affectedhistological structure in liver of pregnant and filial mice.The liver of pregnant andfilial mice at the different developmental stages were damaged in different degrees byephedrine, hepatic sinusoid expand,endotheliocyte fall off, hepatic lobule limitdisorder, with the increase of the dosages of ephedrine and the extension of time,hepatic plate atrophy, hepatic tissue puff, cell cord break section, hepatic plate analosis, livercell confused and disorderly, hepatic sinusoid obviously expand.Compared with theephedrine group, hepatic lobule limit orderly,hepatic sinusoid expand was not obviousand endotheliocyte fall off were not seen of the histological structure in the liver ofthe developing pregnant and filial mice of the traditional Chinese herbal medicinegroup.4. Expression of c-fos protein and TGF-β1in liver was influenced by ephedrineand the traditional Chinese medicine compound,the positive expression of c-fosprotein and TGF-β1in liver of filial mice obviously increased (P <0.01or P <0.05)with injection6.0g/L ephedrine than that of the control at the each developmentalperiod,and the positive expression were stronger with the consistence of ephedrineincreasing. Compared with the6.0g/L ephedrine group,the positive expression ofc-Fos protein and TGF-β1decreased (P <0.05or P <0.01) gradually in the liver offilial mice of the traditional Chinese medicine compound.ConclusionRegular doses of ephedrine can make pregnant mice have tolerability andaddiction,damage the antioxidant enzymes system,cause damage of the histologicalstructure in liver,lipid peroxidation product and free radicals accumulation,cause liverlipid peroxideation damage in pregnant mice.After intraperitoneal injection ofephedrine pregnant mice, ephedrine and metabolin of ephedrine may get into liver offilial mice through placental membrane or breast milk. Ephedrine affected the antioxidant enzymes activity in liver of pregnant and filial mice,there was a great inhibition forthe T-AOC,GST activity of ephedrine groups, but content of MDA evidentlyincreased. The liver of filial mice at the different developmental stages were damagedin different degrees by ephedrine.At the same time,renal tissue TGF-β1and c-Fosprotein expression strength increases,the cell apoptosis degree enhance. Thetraditional Chinese medicine compound contains panaxsaponin,ferulaic acid,glycyrrhizic acid active constituents, these active constituents have obviousantioxidant effect, reduce the lipid peroxidation,hydrogen peroxide super oxygen freeradicals hydroxyl radicals peroxide and the nitro severe clear role. Rhynchophyllineremove location preference of phenyl propyl amine, adjusted the content5-HT、dopamine incerebrum.So the pregnant and filial mice of test index and histological structure have a less damage ininjected ephedrine after irrigation the traditional Chinese medicine compound, antioxidantenzymes activity increases,lipid peroxidation levels was lower, liver tissue TGF-β1and c-Fosprotein expression intensity decreased.That traditional Chinese medicine compound hasgood protect for liver of pregnant and filial mice of ephedrine damage.
