Objective: Comparing the changing of index, which is related to the damage of brain and liver by using different doses of ephedrine after ischemia/reperfusion injury. Exploring whether ephedrine will produce side effects about brain and liver among the dose used in our research .Then to lay a more solid theoretical foundation for the clinical application of ephedrine.Methods: 96 Sprague-Dawley(SD) rats, weighting 220-250g,were used to prepare left middle cerebral artery occlusion (MCAO)models by using modified Longa ligation. Divide the rats into 4 groups randomly: ephedrine A group,ephedrine B group, ephedrine C group and natural recovery group(D). Select 4 times(1st week,2nd week,3rd week,4th week)after the operation to observe. The AST value and pathological section of liver reflect the situation of brain damage, and the AKP value reflect the degree of cholestasis. Expression intensity of brain-derived neurotrophic factor (BDNF) in DG area of hippocampus and pathological section of the brain reflect the situation of brain damage.Results: (1) The determination of AST and AKP showed that all the measured values of medication groups were higher than that of natural recovery group, while ephedrine A group increased the most. Furthermore, all the measured value of medication groups reached a peak value at the 3rd week, then began to fall back at the 4th week; (2) The expression intensity of BDNF in DG area of hippocampus showed that all the medication groups were stronger than natural recovery group, and increased gradually after operation, with a peak value at the 3rd week, then dropped down at the 4th week. Furthermore, the ephedrine A group with the largest change of all;(3)The pathological section results: in all of the medication groups, we can see mild cell edema and hyperemia in the right side of brain, and cell swelling and vacuolar degeneration of liver.Conclusion: Ephedrine has brain and liver side effects when it is used in cerebral ischemia model. There must be correlation between side effects and dose, the greater the dose is given, the more obvious damage it comes. The injury of brain and liver is transient, which can be recovered when discontinuation of treatment.
