Objective:To explore the poison effects of ephedrine and protective effects of lily soup on lung in mice injured by ephedrine,increasing doses of intraperitoneal injection of ephedrine and intragastric administration of lily soup were given to the mice.And some indices were measured,including body weight,spleen index,thymus index,lung index, the activities of plasma MPO and antioxidant enzymes, histological structure and the expression of specific protein related in lung.Methods:90 age of 10 days Kunming mice were chose and randomly divided into 3groups:control group,ephedrine group and lily group.Ephedrine group and lily group were injected escalation doses 2.0 g/L,3.0 g/L and 4.0 g/L of ephedrine(0.2 ml).Then lily group was respectively intragastric administration with 0.2 ml lily soup.Ephedrine group and control group were intragastric with the same amount of saline.And control group was injected of the same amount of saline.The above were operated for 15 days twice a day.The mice in every group were injected for 5,10,and 15 days and were killed to get the weight of body,spleen,thymus and lung,to calculate the viscera indexs.Colorimetry was used to check the activity of MPO in plasma and SOD,CAT and GSH-Px in lung as well as the content of MDA.In addition, the method of paraffin embedding and hematoxylin eosin were used to observe the change of histological structure. Immunohistochemistry was used to detect the expression of Caspase-3,c-Fos,Bax and NF-?B in lung.Results:1.The effect of lily soup on body weight and visceral index in mice injured by ephedrine.Compared with control group,the body weight,spleen index and thymus index in the mice of ephedrine group were reduced and lung index was rised(P<0.01 or P<0.05).Compared with ephedrine group,the body weight,spleen index and thymus index in mice of lily group were rised and lung index was reduced(P<0.05).2.The effect of lily soup on activities of antioxidant enzymes in mice injured by ephedrine.Compared with control group,the activities of SOD,CAT and GSH-Px of ephedrine group were higher temporarily at the 5th treatment day,while significantlylower at the 10 th and 15 th treatment days(P<0.01 or P<0.05).MDA content reduced temporarily at the 5th day, while significantly increased at the 10 th and 15 th day(P<0.01 or P<0.05).Compared with ephedrine group,the activities of SOD,CAT and GSH-Px significantly increased and MDA content reduced at the 10 th and 15 th days(P<0.01 or P<0.05) in lily group.3.The effect of lily soup on lung histological structure in mice injured by ephedrine.The lung histological structure in control group was complete and every level of bronchial were seen clearly,while histopathologic damage in the lung of ephedrine group was obvious. In part, the alveolar structure were disorder and pithelial cells were fall off. The lung histopathologic damage in lily group was lighter.Compared with control group,diameter and thickness of the alveolar septum in ephedrine group increased and radical alveolar counts reduced.And the difference was significant at the 10 th and 15 th treatment days(P<0.01). Compared with ephedrine group,the alveolar diameter and thickness of the alveolar septum reduced and radical alveolar counts increased in lily group.And the difference was significant at the 10 th and 15 th treatment days(P<0.01 or P<0.05).4.The effect of lily soup on activities of plasma MPO in mice injured by ephedrine.Compared with control group,the activity of plasma MPO in ephedrine group were rised(P<0.01 or P<0.05).While compared with ephedrine group,lily group were reduced and the difference was significant at the 15 th treatment day(P<0.05).5.The effect of lily soup on expression of protein(including Caspase-3,c-Fos,Bax and NF-?B)in mice lung injured by ephedrine.Compared with control group,stained color and average optical density of Caspase-3, c-Fos, Bax and NF-?B increased in ephedrine group(P<0.01 or P<0.05).Compared with ephedrine group,the stained color and the average optical density reduced in lily group(P<0.01 or P<0.05).Conclusion:1.Ephedrine had an effect on the body weight and visceral index in mice.Ephedrine increased the excitability and activity of mice.It could disorder the normal development of tissues and induced inflammatory reaction in mice.And theimmune function was reduced.2.Ephedrine had an effect on the activities of antioxidant enzymes of mice lung. Low concentration of ephedrine induced the body immune defenses in mice.The temporary increases in antioxidant enzymes activity and decrease in MDA content induced by the improvement of the genes expression of antioxidant enzymes and the reduction of MDA accumulation in the lung. With the injected-time and concentration of ephedrine increasing, the process of synthesis of antioxidant enzymes were inhibited and the level of free radical was rising.Then the antioxidant enzyme system was damaged.3.Ephedrine had an effect on the lung histological structure in mice. After ephedrine was injected into mice,the number of alveolar in unit area and lung breath area were decreased.Then the lung of oxygen utilization slowed downs.At last the function of the lung was influenced.4.Ephedrine had an effect on the activity of plasma MPO and the expression of Caspase-3,c-Fos,Bax and NF-?B in lung.Ephedrine increased the plasma MPO activity and the expression of Caspase-3?c-Fos?Bax and NF-?B in mice lung.It suggested that ephedrine may accelerated the damage of lung in mice by increased inflammation reaction and cell apoptosis.5.Lily soup had an protective effect on the damage of mice lung induced by ephedrine.There are a variety of trace elements and active substances in lily.It improves the body immunity,balances the antioxidant enzyme system,regulates the metabolism,improves systemic and pulmonary circulation and inhibits inflammatory reaction and cell apoptosis in damaged tissues.In addition,it has the function of fatigue resistance and central sedation.All the indices of lily group was similar with control group. It suggested that lily soup improved the immune function and antioxidant enzymes activities,decreased the inflammatory reaction and cell apoptosis in lung,then repaired the structure of lung in mice injured by ephedrine.
