| Objective: To establish the rat’s middle left tibial fracture, observe the effect of Osx,TGF-β1expression during the healing process with different kidney supplementationmethods, induce osteoblast differentiation and provide certain theoretical basis for promotingfracture healing.Methods:96wistar rats (250+20g, SPF) were selected and randomly divided distilledwater group (A), Bushen Zhuangjin decoction group (B), Zuogui pill group (C) and Youguipill group (D) with24rats in each group. The rat’s middle left tibial fracture were obtained bythree point bending method and fixed small splint (2cm×0.25cm) after the surgery. After6himplantation,8rats in each group were treated, anesthetized abdominally and observed thecurative differences of different kidney supplementation methods with X-ray and callusquantitatively analysed1,2,4weeks after operation, then bone specimens were collected andplaced in4%paraformaldehyde after sacrificed; produce paraffin sections, observe theexpression of Osx, TGF-β1of each group by immunohistochemical methods rats anddetermine the average optical density value (MOD) with ImagePro plus6Image analysissoftware. All the data were analyzed by SPSS17.0.Results: The X-ray imaging of the end of fracture had no significant difference amongB, C, D treatment group and distilled water group the first week after surgery, the fracturehealing of B, C, D treatment group were faster than distilled water group, B group fasterthan C, D group, C group faster than group D after2,4weeks. The callus of the end offracture were quantitatively analysed, there are no statistical significance in each group thefirst week after surgery, B group is significantly higher than that of A group (P <0.01), C, Dgroup higher than that of A group (P <0.05), B group higher than that of C, D group (P <0.05) and C group higher than that of D group (P <0.05). Immunohistochemical resultsshowed the expression of Osx, TGF-β1increased gradually from the1st week to the2ndweek, the decreased expression after the4th week in each group. The average opticaldensity value (MOD) of positive cells of B, C, D group is significantly higher than group A(P <0.05), B group higher than C, D group (P <0.05), group C higher than D group (P < 0.05) after1st,2nd,4th week, positive expression peaked in2nd week.Conclusion: The results show that Bushen Zhuangjin decoction, Zuogui pill and Youguipill can enhance the expression of Osx, TGF-β1and the expression in Bushen Zhuangjindecoction and Yougui pill group are significantly higher than that Zuogui pill group. BushenZhuangjin decoction favor the formation of a cartilage callus of local fracture and promotehealing of the fracture more than Zuogui pill and Yougui pill. |
