Objective: To observe the rapeutic effect of Guijiashutong Capsule on experimentaldysmenorrhea rats and to explore its mechanism of action. Through preparation of dysmenorrheamodel rats and rats uterine smooth muscle strips. To explore the efficacy of Guijiashutongcapsule in the treatment of primary dysmenorrhea foundations, and verify the effectiveness of thetreatment of dysmenorrheal,through the rat anti-inflammatory and analgesic experiments.Methods:(1)The effection of Guijiashutong Capsule on normal uterine smooth muscle contraction andon oxytocin induced uterine smooth muscle contraction:8only normal adult unfertilized femalerats were injected diethylstilbestrol0.5mg/kg before the experiment for5consecutive days.then,by cervical dislocation the rats were sacrificed, shearing of about2cm long uterine which isconnected with the force transducer,in the experiment.①First, to record a normal curve followedby adding different doses of the drug of0.2ml each time, and to observe uterine smooth muscle ofrats in each average tension drug within10min,and frequency of contraction(/l0min).②First toorded a normal curve, adding oxytocin0.lu, then to add different doses of the drugof0.3ml eachtime, and to observe uterine smooth muscle of rats in each average tension within10min, andfrequency of contraction (/10min).(2)Experimental research of Guijiashutong on dysmenorrhea model rats:70female ratswhich were randomly divided into7groups including blank control group、model control group、positive medicine control group、positive control group、Chinese medicine Guijiashutong capsulelow,medium and high dose groups,10rats in each group. After Continuous injection ofsubcutaneous diethylstilbestrol for10d, the dysmenorrhea model rats were establish. From thebeginning of5d, administered daily (or distilled water) for7consecutive days. Groups of animalswere intraperitoneally injected oxytocin2u/only at the end of each animal of administration ofdiethylstilbestrol (distilled water) after24and1hour and to observe Incidence of animal bodytorsion and the incubation period and frequency. After30min,to strip uterus of rat according to ratprostaglandin F2ɑ alpha elisa kit rat prostaglandin and PGE2elisa kit for PGF2ɑ、PGE2contentdetection.(3)Experimental study on anti inflammatory: Guijiashutong capsule effects on egg whiteinduced rat paw edema; Guijiashutong capsule of the influence on cotton ball granuloma of rats. (4)Experimental study on analgesic: Guijiashutong capsule influence on heat induced pain inrats;t Guijiashutong capsule of the influence on the pain caused by acetic acid.Results:(1)Guijiashutong capsule impact on normal rat uterine smooth muscle results suggest thathigh agent of Guijiashutong capsule can reduce the normal rat uterine smooth muscle in vitroaverage tension with significant difference(P<0.05); agent of Guijia Shutong Capsule can reducethe normal rat uterine smooth muscle in vitro the contraction frequency with a significantdifference (P<0.05). Tips on the contraction of uterine smooth muscle frominduced by oxytocinin rats results is high doses of Guijiashutong capsule can antagonize strong contracti of isolted ratuterine smooth muscle and reducing the average tension within10min after administration with avery significant difference (P<0.01); Guijiashutong capsule in high dose, low, can reduce thefrequency of contraction ofuterine smooth muscle from the body with a significant difference(P<0.01).(2)effects of Guijiashutong capsule on pain writhing response of dysmenorrhea model ratinduced by oxytocin is high dose group of Guijiashutong capsule, Yueyueshu and aspirin groupcan recuded writhing latency and the writhing times of dysmenorrhea rat within30min with avery significant difference (P<0.01). Tip on PGF2ɑand PGE2content of dysmenorrhea rat uterus ishigh dose of Guijia Shutong Capsule, Yueyueshu group, aspirin group can reduce the PGF2alphainendometrium of rats with a significant difference (P<0.05); high dose group of Guijia ShutongCapsule, Yueyueshu group, aspirin group can increase the content of PGE2in endometrium ofrats with a significant difference(P<0.05); high dose group of Guijia Shutong Capsule,Yueyueshu group, and aspirin group can reduce the PGF2ɑ/PGE2with a verysignificant difference(P<0.01).(3)Effects of Guijiashutong capsule on the rat foot swelling induced by egg white suggestedthat low, middle and high dose group of Guijiashutong capsule and aspirin group can inhibitinflammation of rat foot swelling. Effects on rat granuloma suggests low, medium, high dosegroup of Guijiashutong capsule and dexamethasone group can reduce the rat granulomacoefficient.(4)Effect of Guijiashutong capsule on pain in mice incude by thermal stimulation suggestthat after30min aspirin group can raise the pain threshold value; after60min, low, middle andhigh dose of Guijiashutong capsule and aspirin group can raise the pain threshold value; after120min high dose group of Guijiashutong capsule and aspirin group can raise the pain thresholdvalue. The influence of Guijiashutong capsule on the number of rats writhing suggest that low, medium, high dose group of Guijiashutong capsule and aspirin has obvious inhibitory effect onmice writhing induced by acid.Conclusion:(1)Guijia Shutong Capsule has obvious inhibiting action on uterine smooth musclecontraction of rats with normal uterine and on uterine muscle contraction induced by oxytocin.(2)Guijia Shutong Capsule has obvious inhibitory effect on writhing reaction ofdysmenorrhea model rats. It can reduce the content of PGF2ɑof dysmenorrhea model rats andincrease the content of PGE2in the uterus and reduce the PGF2ɑ/PGE2.(3)Guijia Shutong Capsule has good anti-inflammation and analgesic action.
