Study On The Expression In Urine Metabolite Profiling Of RA Rabbits By Warm-reinforcing Needling

Objective: Using metabolomics technology to research the function of warm-reinforcingneedling on abnormal metabolism and to explore the pathogenesis in rheumatoid arthritis(RA) rabbits, exploring the mechanism and specificity effects of warm-reinforcing needling intreating RA.Providing the experimental basis for selecting the best acupuncture method fortreating RA.Methods: By SPSS19.0software,30rabbits were randomly divided into5groups:normal group, model group, reinforcing reducing group, twisting reinforcing method group,warm-reinforcing needling group,6rabbits in each group. Except the normal control group,the remaining4groups with ovalbumin induced by the RA model, second days after themodeling, the normal control group and model group grasping method just like otheracupuncture group and fixed1times, each time30min, daily, a total of7days; reinforcingreducing group: with the size of0.25×40mm Hua Tuo brand acupuncture in Zusanli,application of reinforcing reducing techniques,1times per day,30min, a total of7days;twisting reinforcing method group: Specifications for the0.25×40mm Hua Tuo brandacupuncture in Zusanli, application of twisting reinforcing method1times per day,30min, atotal of7days;warm-reinforcing needling group:specifications for the0.25×40mm Hua Tuobrand acupuncture in Zusanli, application of warm-reinforcing needling,1times per day,30min, a total of7days.7days later, the urine of every rabbit been collected and used inmetabonomics analysis, analysis was made with the simca-p11.5software PCA, PLS-DA,draw the corresponding score chart.Results: Established the RA model of rabbit metabolites based on LC-Q-TOF-MSspectrum, the model group and normal control group were compared and analyzed, after therelevant database comparison and identification, obtained25differential metabolism offoreign body, after PLS-DA analysis, and found that all the acupuncture group were able tomake the metabolite profile normal to deviate from the model group, and the warmreinforcing needling group was the best.25differences in the material were identified, andprimarily involved in energy metabolism, amino acids, purine, pyrimidine and unsaturatedfatty acids metabolisms. Conclusions: The disorders of energy metabolism, amino acid metabolism, purinemetabolism, pyrimidine metabolism and fatty acid metabolism may be related to thepathogenesis of RA. Warm reinforcing Needling Treatment of RA make the urine metaboliteprofiling control is better than twisting reinforcing method and reinforcing reducing.Warm-reinforcing needling could reverse the pathological process of RA through partlyregulating the disturbed energy metabolism, amino acid metabolism, fatty acid metabolism.
