Objective: The aim of this study was to identify AnnexinA2as candidate biomarkerin candidate biomarkers for esophageal squamous cell carcinoma (ESCC) Of Kazakh aswell as to reveal the potential role of AnnexinA2on ESCC cell line Eca109.Methods: AnnexinA2’s expression was detected by immunohistochemistry inparaffin-embedded sections from77patients with ESCC of Kazakh. ESCC cell lineEca109cells were transfected with full length cDNA of AnnexinA2eukaryotic expressionvector pCMV-XL5-AnnexinA2. The expression of AnnexinA2was detected by qRT-PCRand Western blot on mRNA and protein level, respectively, proliferation was examined byMTT assay, migration capacity was evaluated by wound healing assay. In addition, the cellapoptosis and cell-cycle was analyzed by flow cytometry.Results: The expression level of AnnexinA2protein was significantly correlated withpatients’ degree of differentiation status and the depth of invasion. AnnexinA2expressionwas successfully and significantly increased and there was a significant decrease in theproliferation, migration and invasion capability of transfected Eca109. In addition,up-regulating AnnexinA2partly caused Eca109cell cycle arrest at the G2phase, and didnot leads to cell apoptosis.Conclusions: our findings suggest that AnnexinA2could be a useful marker for evaluationof ESCC progression and played an important role in the tumorigenesis of Kazakh’sESCC.
