Objective To observe the effects of Yuyang decoction(YD)on theexpressions of TFF2and TFF3in Acetic Acid-induced Chronic Gastric UlcerRats, and to explore the role of YD in the progress of ulcer healing. Methods60SD rats were randomly divided into normal group, model group, high-dose YDgroup, mid-dose YD group, low-dose YD group and group Rabeprazole. The ratmodel of chronic gastric ulcer was established by glacial acetic acid,morphological changes of gastric tissue were observed under microscopyby HE stain, the expressions of TFF2and TFF3in gastric antrum were assayedby immunohistochemistry(IHC)and reverse transcription/polymerase chainreaction respectively(RT-PCR). The average optical density (OD) and the PCRproduct were estimated by Image-Pro Plus6.0software. Results The GUI inDrug intervention groups and normal group were decreased than that in modelgroup (P<0.01或P<0.05).The GUI in high-dose and mid-dose YD group(6.88,3.44g/kg) was significantly lower than that in group Rabeprazole (P<0.01或P<0.05).compared with the low-dose YD group(1.72g/kg), the GUI in groupRabeprazole was lower, but the results were not statisticallysignificant(P>0.05).Under the microscope, normal group shows normal gastrictissue with smooth and pink gastric mucosa, neat and clear duplicature, noerosion and ulcer. The microstructure of gastric tissue of rats in model groupshows ulcer formation and mass inflammatory cells, necrotic tissue in mucosal surface layer, granulation tissue proliferation in submucosal layer. The musclelayer and serosa of rats in model group was destroyed completely. Comparedwith model group, the microstructure of gastric tissue of rats in Drugintervention groups were better obviously, the thickness of regenerative mucosaraised, the numbers of inflammatory cells surrounding ulcer decreased obviously,and the recovering of serosa layer accelerated. IHC demonstrated that theexpression of TFF2and TFF3in model group and drug intervention groups wereincreased than that of normal group(P<0.01或P<0.05). The expression of TFF2and TFF3in high-dose and mid-dose YD group was significantly increasedcompared with model group and group Rabeprazole(P<0.01或P<0.05).Thedifferences among the low-dose group、model group and group Rabeprazole,were not statistically significant (P>0.05). TFF2mRNA、TFF3mRNA werescarcely detected in antrum of rats of normal group. The TFF2mRNA andTFF3mRNA in high-dose and mid-dose YD group was increased compared withmodel group and group Rabeprazol(eP<0.01或P<0.05).The differences amongthe low-dose group、model group and group Rabeprazole, were not statisticallysignificant (P>0.05).Conclusion Yuyang decoction can improve theexpressions of TFF2and TFF3to protect the gastric mucosa of rats, toaccelerate ulcer’s recovering, and to enhance the quality of healing rate.
